摘 要: | 目的为了对微生物检测基因芯片杂交荧光信号进行有效判读。方法通过计算预测结合自由能的方法,充分考虑寡核苷酸探针和目标16S rRNA所有可能的碱基配对,分析计算杂交反应自由能与杂交荧光强度的关系。结果特异性与非特异性杂交反应自由能的差和两种杂交情况下荧光强度的比值具有较高的相关性。结论阳性对照辅助的多阈值微生物检测芯片信号判读系统是可实现的。

Abstract: | Objective To judge fluorescence signal of microorganism detection gene microarray efficiently. Methods Based on computational methods for free energy prediction, taking all possible base pairs between oligonucleotide probe and target 16S rRNA into consideration, relationships between free energy of hybridization and fluorescence signal intensity were analyzed. Results Difference of free energy between specific and no.specific hybridization is highly related to the ratio of fluorescence signal intensity received in these two hybridizations. Conclusion Multiple-threshold judgement system for microorganism detection microarray based on positive control is available. |
