不同时窗针刺对窒息脑瘫幼鼠脑组织bFGF表达的影响

摘    要:目的:探讨针刺在不同时期介入对窒息脑瘫幼鼠的治疗作用及其神经生化机制。方法:7日龄新生幼鼠28只随机分为4组:模型+早期针刺组(A组),模型+晚期针刺组(B组),模型组(C组)。假手术对照组(D组)。A、B组分别于造模后24h、8d开始针刺;动态观察其行为学改变,于21d时处死,测定脑组织碱性成纤维细胞生长因子(bFGF)的表达情况。结果:造模后幼鼠前肢功能明显下降。至21d时,A、B、C3组前肢功能均显著改善。A、B组基本接近正常水平,C组差于D组。造模后21d,D组幼鼠在皮层、纹状体、海马仅有轻微的bFGF免疫反应,C组幼鼠见稍增强的bFGF阳性表达,而A、B组均见较多量的bFGF强阳性表达,尤以A组为明显,各组间差异有显著性意义(P〈0.001)。结论:针刺早期介入对脑瘫的治疗有肯定作用。增强的bFGF长时程阳性表达,可能是针刺治疗窒息性脑瘫的重要机制之一。

Abstract:Objective: To observe the effects and neural biochemical mechanism of acupuncture at different time-windows on hypoxic-ischemic cerebral palsy rats. Methods: Neonatal SD rats were randomized into acupuncture group-1 (group A, acupuncture was given at 24th h after operation), acupuncture group-2 (group B, acupuncture was given at 8th day after operation), group C and group D. The forepaw-touching test (adhesive plaster tearing-off test) was done and the expression of basic fibroblast growth factor (bFGF) in the brain was assayed by using immunohistochemical method. Results: The function of the rat's forelimbs were obviously descended after operation. Till the 21st day, the function of the rats' forelimbs in group A, B and C were apparently improved, and the rats' function in group A and B were almost near to normal standerd, but rats in the C group were inferior to these of the D group. Twenty-one days after operation, the number of bFGF immunoreaction (IR) strongly-positive neurons in the cerebral cortex, straiate body and hippocampus was most in group A, more in group B and less in group C, and only very weakly IR-positive neurons were found in group D(P<0.001). Conclusion: Acupuncture therapy may be used to treat cerebral palsy at the early stage. Acupuncture can up-regulate the long-term expression of bFGF in the brain, which may contribute to the effect of acupuncture in improving cerebral palsy.
