银杏内酯B协同地塞米松对小鼠哮喘模型气道炎症和辅助T细胞亚群的影响

摘    要:目的:探讨银杏内酯B协同地塞米松对支气管哮喘气道变应性炎症及辅助T细胞亚群的作用。方法:40只BALB/c小鼠随机分成正常对照组、哮喘对照组、地塞米松组、银杏内酯B组和联合干预组,每组8只。除正常对照组外,其余4组均造成哮喘模型,地塞米松组、银杏内酯B组和联合干预组给予相应的药物。收集支气管肺泡灌洗液(BALF),行白细胞(WBC)及嗜酸性粒细胞(EOS)计数;应用EUSA法测定BALF中IFN-γ和IL-4水平。结果:BALF中WBC及EOS计数:3个用药组较哮喘对照组明显减少(P〈0.01),联合干预组减少的程度大于地塞米松组、银杏内酯B组(P〈0.05或P〈0.01)。3个用药组小鼠BALF上清中IFN—γ和IL-4水平与哮喘对照组比较,银杏内酯B组、地塞米松组和联合干预组BALF上清中IFN-γ和IL-4水平分别增高和降低(P〈0.05或P〈0.01),而联合干预组增高和降低的程度大于地塞米松组、银杏内酯B组(P〈0.05或P〈0.01)。结论:银杏内酯B能减轻小鼠哮喘模型BALF中炎性细胞浸润,并减少IL-4的水平;银杏内酯B与激素具有协同抗炎作用;同时具有更好的调节Th亚群失衡的作用。

Effects of ginkgolide B coordinated with dexainethasone on allergic airway inflammation and T helper cell subsets in asthmatic rats

Abstract:Objective:To investigate the effect of ginkgolide B(DB) coordinated with dexamethasone(DXM) on allergic airway inflammation and T helper cell subsets in asthmatic rats. Methods:Forty BALB/c rats were randomizedly divided into 5 groups equally:the normal group,the asthma model group, the DXM group ,the GB group and the DXM plus GB group,they were treated with correspondant medicines. The counts of WBC and eosinophil(EOS) in bronchoalveolar lavage fluid (BALF) were estimated and ELISA was used to determine the concentrations of IFN-γ and IL-4 in BALF. Results:Compared with the asthma model greup,EOS and WBC count and IL-4 level in BALF were significantly lower in the three treated groups(P〈0.01 ),particularly in the DXM plus GB group,showing a significant difference as compared with the other two groups (P〈0.05 or P〈0.01 );whereas IFN-γ levels were increased in the DXM plus GB group, they had significant difference as compared with the other two groups (P〈0.05 or P〈0.01 ). Conclusion:Gingkgolide B combined with dexamethasone has synergistic anti-inflammatory effects on airway inflammation, and might partly reverse T helper cell subsets better in rats with asthma.
