酒精依赖大鼠模型建立

目的:建立酒精依赖大鼠模型。方法:给大鼠饮含6%(v/v)酒精水溶液28d。观察饮酒期间大鼠动机活性、撤除酒精后的戒断综合征及抑郁症状。结果:饮酒组大鼠动机活性出现一适应性变化过程,即在饮酒7、14d大鼠探究行为(站立次数)和水平动机活度(爬格数)与对照组相比显著降低(P<0·01),而于饮酒28d恢复正常。饮酒组大鼠戒断2-48h戒断症状评分均明显比对照组高,以戒断6h评分最高(16·50±s3·12);酒精戒断24、48h饮酒组大鼠在强迫游泳实验中绝对不动时间与对照组比较显著延长,至戒断72h仍未恢复。结论:大鼠连续饮用含酒精6%(v/v)的水溶液,能建立可重复性、稳定的酒精依赖动物模型。

ESTABLISHMENT OF RAT ALCOHOL DEPENDENT MODEL

Objective: To establish the model of ethanol dependent rats. Methods: Ethanol was administered in drinking water at the concentration of 6% v/v, for 28 days. The effects of chronic alcohol intake on the locomotor activity were investigated in the open-field. Alcohol withdrawal syndromes were evaluated after alcohol withdrawal. The symptoms of depression were examined by forced swim test (FST). Results: Rats consuming a liquid containing 6% ethanol for 7, 14 days exhibited a significant decrease ambulation activity,accompanied by a reduced frequency of explorative rearing in an open-field task, whereas presumed adaptation to the neurologic effects of alcohol was observed on day 28. The withdrawal syndrome scores of ethanol-fed rats were obviously higher than those of control rats during 2-48 h after alcohol withdrawal, the highest score which occurred at 6 h after ethanol withdrawal was 16.50±s 3.12. In addition, alcohol withdrawal could be accompanied by symptoms of depression, and the immobility of rats was increased in the FST at 24 h of alcohol withdrawal, and the increment remained at 48 h, and 72 h of alcohol withdrawal. Conclusion: A stable, repetitive animal model for alcohol dependence can be established by giving long-term intake of ethanol solution to rats.
