弱精不育患者与正常生育能力者精浆蛋白质的双向电泳图谱分析

目的:探讨2-DE技术在人类正常与弱精不育者精浆全蛋白中筛选差异蛋白的应用。方法:先用IPG-DALT技术对12例弱精不育与5例正常精浆蛋白进行分离,银染后,经PDQUest2D比较二者间差异蛋白点。结果:在不育精浆中高丰度存在而正常精浆中低表达的有55个蛋白斑点(3个在正常缺失),在正常精浆中存在而在不育精浆中低表达的有43个(3个在不育缺失)。结论:成功建立了正常与弱精不育精浆蛋白的2-DE图谱并分析表达差异蛋白,为弱精症精浆差异表达蛋白的质谱鉴定奠定了基础。

Differential Analysis of Two-dimensional Gel Electrophoresis Profiles of Seminal Plasma Protein in Human Asthenospermia Seminal Plasma and Normal Seminal Plasma

Objective:To evaluate the application of two-dimensional electrophoresis in the research of differentially expressed proteins in human seminal plasma.Methods:Seminal plasma total proteins were separated from 12 asthenospermia patients and 5 normal healthy men through IPG-DALT.After silver staining,PDQuest 2D image software was used to analyze differential expressed proteins in 2-DE gel.Results:After analyzed,when optical density value between normal and sample was at five,55 proteins spots were expressed highly in asthenospermia seminal plasma but lowly or absence in the normal(3 proteins absent in normal samples),43 protein spots existed in normal healthy men seminal plasma but lowly or absence in the asthenospermia patients(3 proteins absent in infertility samples).Conclusion:The protein profiles of differential expression between the normal seminal plasma and asthenospermia patients were established and some differential proteins were found.The data obtained in this study may help to prepare the ground for further studies on the isolation and identification of differentially expressed proteins in human asthenospermia seminal plasma.
