As2O3对正常免疫力小鼠移植性卵巢肿瘤作用的实验研究

目的：观察As2O3的体内抗卵巢癌效应。方法：采用人卵巢癌COC1细胞系，以正常免疫力的雌性近交系BalB/c小鼠为对象，建立了人卵巢癌的小鼠皮下移植瘤模型；小鼠移植瘤经病理组织学检查及免疫组化法鉴定肿瘤组织的性质及来源；筛选砷剂腹腔注射的剂量并取急性中毒死亡小鼠的脑、肝、肾做病理组织学检查；采用腹腔注射法给药，观察砷剂的体内抗卵巢癌作用；治疗组小鼠的脑、肝、肾做病理组织学检查。结果：经病理组织学及免疫组化法鉴定证实小鼠移植疡为人低分化性上皮性卵巢肿瘤；As2O3可以显著抑制小鼠移植瘤生长；大剂量的砷剂可引起小鼠脑组织及肝组织坏死；治疗剂量(1mg/kg.d和2mg/kg.d)的砷剂对小鼠脑、肝、肾等重要脏器未造成明显损害。结论：As2O3对卵巢癌具有良好的治疗作用。

EFFECTS OF AS2O3 ON TRANSPLANTING OVARIAN CANCER IN THE NORMAL IMMUNITY MOUSE

Objective:To determine the effect of As 2O 3 on ovarian cancer by internal experiment.Methods:We used COC 1 cell line and normal immunity inbreed line BalB/c mice as objects to construct the subcutaneous transplanting ovarian cancer model,and histopathological and immunohistochemical method to confirm the property and source of the transplantation tumor. After selecting the dose of As 2O 3 for abdominal administration, the brain, liver and kidney of poisoning mouse were examined by histopathological method. We observed the effect of As 2O 3 on ovarian cancer after abdominal administration. The brain , liver and kidney of mouse in therapeutic group were examined by histopathological method.Results:Histopathological and immunohistochemical experiment proved that the transplantation tumor was undifferentiated epithelial ovarian cancer; As 2O 3 could significantly inhibit the growth of transplantation tumor;the lethal dose As 2O 3 could cause brain necrosis and liver necrosis; therapeutic dose As 2O 3 (1mg/kg.d and 2mg/kg.d) didn't damage the brain, liver and kidney of mouse.Conclusion:As 2O 3 has significant therapeutic action on ovarian cancer.
