摘    要:目的研究斑蝥酸钠对人肝癌细胞的增殖作用及其对血管内皮生长因子(VEGF)的表达的影响,探讨斑蝥酸钠抗肿瘤的作用机制。方法体外培养人肝癌细胞株(HepG2),用不同浓度的斑蝥酸钠作用于肝癌细胞,采用MTT法观察不同浓度和作用时间下斑蝥酸钠对细胞增殖的影响,采用免疫组化法检测血管内皮生长因子的表达情况。结果在一定浓度范围内斑蝥酸钠对肝癌细胞有明显增殖抑制作用,且降低了VEGF的分泌水平,并呈时间-剂量依赖关系。结论斑蝥酸钠对肝癌细胞的增殖有抑制作用,且减低了细胞内血管内皮生长因子蛋白的表达,表明斑蝥酸钠在抑制肝癌生长的作用机制之一是通过抑制肿瘤血管生成发挥作用的。

Abstract:Objective To study the effects of sodium cantharidate on cell proliferation and VEGF expression in human human hepatocellular carcinoma cells and explore its possible mechanisms of antitumor.Methods Human hepatoma cells HepG2 were cultured in vitro and tread with different concentration of sodium cantharidinate,then cell proliferation was examined by the method of MTT.Immunohistochemistry was applied to examine the protein expression of VEGF.Result In certain concentration range,sodium cantharidate can inhibit cellular proliferation and down-regulated the protein expression of VEGF,the time-dosage dependent relationship existed.Conclusion Sodium cantharidate can inhibit liver cancer cell proliferation,and lower the expression of VEGF protein.Show one of mechanisms of sodium cantharidate inhibiting hepatocellular carcinoma growth is by inhibiting tumor angiogenesis.
