前房不同部位注射复方卡波姆诱导兔慢性青光眼模型的实验研究

摘    要:目的 通过评价前房不同部位注射复方卡波姆建立兔慢性青光眼模型的效果,探讨此模型的最佳注药部位.方法 将20只兔随机分为模型Ⅰ、Ⅱ组,右眼为模型眼,分别用复方卡波姆制作慢性高眼压模型.模型Ⅰ组将复方卡波姆缓慢注射在前房瞳孔区,模型Ⅱ组缓慢注入周边房角.左眼为对照眼,前房内均注入生理盐水.兔眼在造模前测量3日眼压值作为基础眼压,造模后2次/d测量,仔细观察眼压的变化规律及持续时间,并对诱发的高眼压模型观察8周.直接检眼镜观察视盘凹陷,光镜观察视网膜的组织形态学改变,视网膜各层组织厚度变化并进行图像分析,评价2组模型的造模效果.结果 Ⅰ组平均眼压(27.2±1.2)mmHg,持续时间为25~53d,Ⅱ组平均眼压(39.6±2.4)mmHg,持续时间为22~28d.光镜下显示模型眼视乳头凹陷均扩大和加深,2组视网膜神经纤维层均变薄,Ⅱ组比Ⅰ组变薄更明显,两组间差异有统计学意义(P<0.05).结论 复方卡波姆诱导兔慢性青光眼模型时注射在前房瞳孔区比周边房角所建立的模型效果更理想,更易于控制和实用.

Experimental study of carbomer glaucoma model in rabbits by injecting different location in anterior chamber
Abstract:Objective To evaluate the effect of carbomer glaucoma model in rabbits by injecting dif-ferent location in anterior chamber,to investigate the best injection rite.Methods Twenty rabbits were divided into two groups:group Ⅰ and Ⅱ.The fight eyes are model eyes,injecting slowly the pupil center in anterior chamber(Ⅰ)and injecting the perimeter in anterior chamber(Ⅱ)to induce carbomer glaucoma.The left eyes injecting NS in anterior chamber as control.The intraocular pressure(IOP)was monitored with a Tono-pen XL tonometer three days prior to injection and 2 times per day after injection.The basic IOP in every group served as control.After 8 weeks,the results were studied.The optic disk and the thickness of retina were observed and measured.Results The mean IOP levels were(27.2±1.2)mmHg, sustained 25~53 days,and(39.6±2.4) mmHg,22~28 days in group Ⅰ and Ⅱ.The cup of the optic disk was changed significantly.The retinal nerve fi-bre layer were thinning in group Ⅰ and Ⅱ.And there was significant difference between twe models.Conclu-sion Experimental glaucoma model induced by carbomer is easy to be carded out.And the method by inject-ing slowly the pupil center in anterior chamber is more effective than the perimeter in anterior chamber.
