Rapid Latex Agglutination Test for Detection of Staphylococcal Enterotoxins

Abstract:Staphylococcal enterotoxins (SE) play an important role in the pathogenesis of human infection of Staphylococcus aureus. After comparing various sensitzing effects with dif- ferent latex particles and different sensitizing methods,we attempted to develop a reversed passive latex agglutination method (RPLA) for rapid detection of SEA, SEB, SEC and SED. Examined by the method, all the 19 ref- erence enterotoxigenic staphylococcus aureus strains and I non-toxigenic strain showed positive and negative, respectively. We also studied the production of SE by 87 staphylo- coccus aureus strains isolated from hospitals in Beijing, Xian and Jinan. The incidence of entero-toxigenic strains was 85.1%(74/87), which was higher than theincidence of 65.5 (57/87)detected by the double immuno-geld- iffusion (DIGD) and was close to the incidence of 77.0%(67/87)tested by the reversed pas- sive hemagglutination test (RPHA). Of en- terotoxigenic strains studied by the RPLA, ablend of SEA + SEB + SEC + SED was pro- duced by the maximum number of the sterains, and by DIGD and RPHA, SEA(34/ 57, 24/67) was predominant, respectively. The method was shown to be sensitive(mini- mum SE detectabl limit was 6~12ng/ml), highly specific (no agglutination occured with Cholera toxin and cultures of E. coli, Shigel- la, Bacillas subtilis and pseudomonas aerogi- nosa)and rapid (took 5~7h for incubation at room temperature). These showed the RPLA is fully reliable for use.
