辐射后造血细胞对造血因子增殖反应性变化的研究

摘    要:目的:深化造血细胞辐射损伤机理的认识,并为造血因子的合理应用提供理论指导。方法:应用MTT法观察4种造血因子依赖细胞株于照射后不同时间对6种造血因子的增殖反应性变化。结果:造血因子依赖细胞株受照后对造血因子的增殖反应性降低,表现为细胞因子ED50值的增加及最大增殖幅度的降低,照射剂量越大,反应性降低越明显,恢复所需时间越长。结论:增殖反应性降低可能是造血因子临床应用时剂量偏大的原因之一,并提示在造

Abstract:Objective: To deepen the knowledge of the mechanism of radiation injury of hematopoietic cells (HCs) and to provide theoretical direction for reasonable administration of hematopoietic growth factors (HGFs). Methods: The changes in growth responsiveness of four HGFdependent cell lines to six HGFs were observed using MTT methods. Results: The growth responsiveness of HCs to HGFs decreased after irradiation in a dosedependent manner and showed that the ED50 of HGFs was increased and the maximal range of growth of HCs was reduced. The higher the radiation dose, the longer the time needed for irradiated HCs to recover. Conclusion: The reduction of growthresponsiveness of HCs to HGFs after irradiation might be one of the reasons for clinical use of HGFs at rather high doses. It is also suggested that there be somewhat obstruction of cytokine receptor signal transduction in the course of radiation injury of HCs.
