长期低浓度3,3’二吲哚甲烷处理鼻咽癌细胞后对裸鼠移植瘤增殖及转移的抑制作用

摘    要:目的低浓度3,3I_二吲哚甲烷(3,3'-Diindolylmethane,DIM)处理鼻咽癌细胞5.8F和CNE-2后,鼻咽癌细胞的增殖及转移能力的抑制作用及其机制。方法浓度为20ImaoVLDIM处理鼻咽癌细胞株5.8F和CNE-2一个月,48只BALB/C裸小鼠随机分为CNE-2/DIM(实验组)及5-8F/DIM(实验组)、CNE-2(对照组)及5-8F(对照组)4组,实验组于裸鼠背部靠腋窝皮下接种经过DIM处理的人鼻咽癌细胞CNE-2/DIM和5-8F/DIM,对照组接种未经DIM处理的鼻咽癌细胞,给予常规裸鼠饲料,每周记录移植瘤的体积,于接种第8周处死动物,并留取裸鼠移植瘤及腋窝和腹股沟的淋巴结组织,检验其PCNA、Ki-67及E.Cadherin、Vimentin的表达。结果接种鼻咽癌细胞8周后,实验组移植瘤体积明显小于对照组,差异具有统计学意义(P〈0.05)。对照组移植瘤成功率及转移率明显大于实验组中PCNA、Ki-67及Vimentin的相对表达强度低于对照组(P〈0.01),而E-cadh丽n的相对表达强度明显高于对照组(P〈0.01)。结论低浓度DIM长期处理鼻咽癌细胞后具有抑制鼻咽癌裸鼠移植瘤增殖和转移的作用,其机制可能与PCNA、Ki-67、Vimentin及E.Cadherin相关。

Abstract:Objective To explore the effects of 3,Y-Diindolyimethane (DIM) on prolifexafion and metastasis of nasopharyngeal earcinoma(NPC) cells 5-8 F and CNE-2 by xcnotransplanted nasopharyngeal carcinoma which treatment with low concentration of DIM in nude mice. Methods The concentration of 20 umol/L DIM managed in NPC cell lines 5-8 F and CNE-2 in a month. 48 BALB/C mice were randomly divided into CNE-2/DIM (experimental group) and 5-8 F/DIM (experimental group), CNE-2 (control group) and 5-8 F (control group). In the experimental group were inoculated subcutaneously in nude mice by axillary back with CNE-2/DIM and 5-8 F/DIM human NPC cells treated with DIM, control group were inoculated with untreated NPC cells. All mice were fed for conventional nude mice food. We recorded the volume change of transplantable tumor every week. We killed the animal in eighth weeks, and remains the nude mice transplanted tumor and axillary and inguinal lymph node tissue, test the expression of PCNA, Ki-67 and E-Cadherin, Vimenfin. Results In experimental group, the tumor volume was significantly less than the control group, there was a statistically significant difference (P〈0.05), after inoculation of NPC cells 8 weeks. The success rate of transplantation tumor e and the metastasis rate in nude micof the control group was lower than the experimental group. The relative expression intensity of PCNA, Ki-67 and Vimentin in the experimental group were lower than the control group (P〈0.01), and E-Cadhcrin expression intensity was significantly higher than the control group (P〈0.01). Conclusion Long-term and low concentration treatment of DIM in nasopharyngeal carcinoma cells inhibits the proliferation and metastasis of NPC in nude mice, and its mechanism may be related to PCNA, Ki-67, Vimcntin and E-Cadhcrin.
