Gastrokine2在胃癌中的表达及临床意义

摘    要:目的探讨Gastrokine 2在胃癌的发生、发展过程中的作用。方法应用实时荧光定量聚合酶链反应(RT-PCR)和Western blot印迹法检测76例诊断明确的胃癌患者的癌组织及癌旁组织中Gastrokine2的表达,并分析胃癌组织中mRNA和蛋白的表达与临床病理特征之间的关系。结果 Gastrokine 2 mRNA和蛋白在胃癌组织中的表达均低于癌旁组织,差异有统计学意义(P〈0.01)。胃癌组织中Gastrokine 2 mRNA的表达与肿瘤的位置、浸润深度、分化程度、Lauren分型、淋巴结转移及患者的性别和年龄均无关(P〉0.01),而幽门螺杆菌(HP)阳性胃癌患者的癌组织中Gastrokine 2 mRNA表达低于HP阴性者(P〈0.01),Ⅰ期胃癌患者的Gastrokine 2 mRNA表达量高于Ⅱ、Ⅲ、Ⅳ期胃癌患者(P〈0.01)。胃癌组织中Gastrokine 2蛋白的表达与肿瘤的位置、浸润深度、分化程度、Lauren分型、淋巴结转移、TNM分期、幽门螺杆菌感染及患者的性别和年龄均无关(P〉0.01)。结论 Gastrokine 2在胃癌组织中的表达显著降低,它作为一种保护性基因在胃癌的发生和发展中扮演着重要角色,随着胃癌的进展,其抑制肿瘤的作用降低,同时HP感染可能减弱其对胃黏膜的保护作用。

Expression and cUnieal significance of Gastrokine 2 in gastric cancer

Abstract:Objective To clarify the role of Gastrokine 2 in the process of formation and development of gastric cancer. Methods The expressions of Gastrokine 2 in gastric cancer and paracancerous tissues of 76 patients with gastric cancer were detected by real-time fluorescence quantitative polymerase chain reaction (RT-PCR) and Western blot. The relationship between Gastrokine 2 expression and clinico-pathologic characteristics of gastric cancer was analyzed. Results Gastrokine 2 protein and mRNA levels were reduced significantly in gastric cancer tissues compared with controls(P〈0.01). Gastrokine 2 rnRNA level had no remarkable correlation with tumor location, depth of invasion, differentiation, lauren, lymph node metastasis, gender and age(P〉0.01, respectively), rather than TNM stage(P〈0.01). In addition, Gastrokine 2 protein level also had no remark_able correlation with tumor location, depth of invasion, differentiation, lauren, TNM stage, lymph node metastasis, helicobacter pylori(HP) infection gender and age(P〉0.01, respectively). However, HP infection reduced Gaslrokine 2 mRNA levels significantly in gastric cancer tissues compared with controls(P〈0.01). Conclusions The expression levels ofGastrokine 2 gene and protein in the gastric cancer tissues were significantly low. Gastrokine 2 may play a significant role as an anti-oncogene in the process of the formation and development of gastric cancer. Its effect may become weak as the development of tumor and HP infection in gastric cancer patients.
