甲型肝炎病毒血症及体外增殖的实验研究

采用PCR、病毒分离及免疫电镜等技术,以甲型肝炎密切接触者及现症病人为研究对象,研究HAV感染机体后的病毒血症,病毒在血液系统中的消长动态及在人血白细胞和单核巨噬细胞中体外增殖和定位,探讨其与致病机理的关系。结果:机体感染HAV后,从病人血清中直接分离出HAV,外周血白细胞HAV抗原阳性,且在潜伏期中病毒即已侵犯外周血白细胞,抗-HAV-IgM阳性血清中有HAV RNA;并证实HAV可在体外培养的人血单个核细胞及单核巨噬细胞中增殖并传代。

Experimental Study of Hepatitis A virusemia and in vitro Multiplication

A methodology of polymerase chain reaction amplification, virus isolation and immunoelectronic microscopy were used to study the patients of hepatitis A and their close contacts, in order to further investigate hepatitis A virus - caused virusemia, the dynamics of the viruses in blood system, and the in vitro multiplication and location of the virus in the peripheral leukocytes and mononu-clear macrophage, and finally to reveal the pathogenesis of the virus. It was shown that after HAV infection, HAV could be isolated directly from serum of the patients, HAAG could be detected in the peripheral leukocytes ,the HAV invaded into peripheral leukocytes at the infection latency, HAV RNA could be detected in the serum immune complex of the patients, and HAV could multplicate in the in vitro cultures of human mononuclerar leukocytes and macrophages.
