染料木黄酮的诱变性研究

摘    要:目的检测和评价染料木黄酮的诱变性,为其安全应用提供毒理学依据。方法采用小鼠淋巴瘤细胞实验(微孔板法)评价染料木黄酮的诱变性,诱变性检测采用3h和24h染毒两种处理方式观察受试物对L5178Y细胞的诱变性。结果在染料木黄酮诱变性检测中,3h染毒时,随着剂量增加,tk位点总突变频率升高,5、10和20μg/mL剂量组tk位点总突变频率与溶剂对照组比较差异有统计学意义(P0.01);24h染毒时,随着剂量增加,tk位点总突变频率升高,与溶剂对照组比较,5、10μg/mL剂量组tk位点总突变频率高于溶剂对照组,差异有统计学意义(P0.01);3h染毒和24h染毒相同剂量组间比较,tk位点总突变频率差异无统计学意义(P0.05)。结论在此实验条件下,染料木黄酮(5~20)μg/mL能诱导L5178Y细胞TK基因突变,并呈良好的剂量反应关系,但3h和24h处理的突变频率差异无统计学意义(P0.05)。

Abstract:Objective To detect and evaluate the mutagenic effects of genistein,and provide basis for its development and application.Methods The mouse lymphoma assay was used in the evaluation of the mutagenic effects of genistein.In the evaluation of mutagenic effect,two kinds of treatment (3h-and 24h-exposure) were used to observe the mutagenic effects.Results In the evaluation of mutagenic effect of genistein,in 3h-exposure treatment,four dosage groups (2.5,5,10 and 20g/ml) were set up.The total mutation frequencies in 5,10 and 20g/ml groups were higher than the solvent control group (P < 0.01);in 24h-exposure treatment,four dose groups (1.25,2.5,5 and 10g/ml) were set up.Compared with the solvent control group the total mutation frequencies in 5,10g/ml dosage groups increased,the difference was statistically significant (P < 0.01);the same dosage groups in 3h-treatment compared with these in 24h-treatment,the total frequencies had no significant difference (P > 0.05).Conclusion In this experimental condition,genistein could induce TK gene mutation in L5178Y cells,and had a good dose-response relationship,without time-response relationship(P >0.05).
