OPNmRNA与BSPmRNA在羊下颌骨牵张成骨过程中的表达

摘    要:目的 观察骨桥蛋白(Osteopontin,OPN)与骨涎蛋白(Bonesialoprotein BSP)在羊下颌骨牵张成骨过程中的表达情况。方法 用自制的牙支抗下颌骨牵引装置,对山羊下颌骨进行牵张,速度为0.25mm/12h,共牵引16天,分别于开始牵引的8、16、32、48天取材,观察颌骨的延长情况;标本常规进行OPN与BSP mRNA的原位杂交反应,并对阳性细胞进行计数。结果 羊下颌骨成功地获得了延长;8天时OPNmRNA少量出现在成纤维样细胞及幼稚的成骨细胞内。之后以成骨细胞内表达为主;阳性细胞数在16天组明显增多,32天组最高,之后减少。BSPmRNA仅见于成骨细胞内,阳性细胞数随着时间延长逐渐增加。结论 OPN与BSPmRNA主要在成骨细胞内表达,与成骨细胞的成熟程度一致,表明它们与牵张成骨过程中新骨的成熟和矿化相关。

Abstract:Objective To observe the expression of osteopontin(OPN) and bone sialoprotein(BSP) mRNA during mandibular distraction osteogenesis in goat model.Methods After a mandibular cortitomy was made between the second and the third premolars bilaterally,tooth-supported distraction devices made by ourselves were cemented onto the second and third premolars.The distraction devices were activated ~0.25mm every 12 hours to advance the anterior segment 8mm in 16 days.The animals were sacrificed on the 8~th ,16~th ,32~nd ,48~th days after activation respectively.Then OPN and BSPmRNA in situ hybridization was conducted,the number of positive cells was counted.Results OPNmRNA was found in fibroblast-like cells and immature osteoblasts at the 8~th days,then was mainly found in osteoblasts.The number of positive cells began to increase at the 16~th days and reached a peak at the 32~nd days,then decreased.BSPmRNA was found in osteoblasts only,the number of positive cells increased with time.Conclusion OPN and BSPmRNA were primarily expressed in osteoblasts,and the positive incidence increased during experimental time,which indicated that they might be connected with the maturation and mineralization of new bone during distraction osteogenesis.
