枸杞多糖对家兔动脉粥样硬化治疗作用的研究

目的：本研究给予动脉粥样硬化（AS）模型动物枸杞多糖（LBP）,通过对α-平滑肌肌动蛋白（α-SMA）和转化生长因子β（TGF-β）在动脉粥样硬化斑块组织中表达的研究及形态学观察,探讨LBP对AS的治疗作用及其作用机制。方法：32只健康雄性新西兰白兔,随机平均分为4组：正常对照组、模型对照组、口服治疗组、注射治疗组。正常对照组喂饲基础饲料,其余各组每天给予高胆固醇饲料喂养10周。10周后,模型对照组改喂饲普通饲料;口服治疗组每天给予基础饲料＋枸杞多糖20 g/kg喂饲;注射治疗组每天给予基础饲料＋枸杞多糖3 mg/kg（注射）。20周末所有实验用家兔使用空气栓塞法处死,各组行主动脉粥样硬化病理学形态观察,并用M asson染色和免疫组织化学染色法对斑块内的病变进行定性定量分析。结果：两种检测方法均可见治疗各组病变较模型对照组明显减轻,以注射组尤为明显,且差异有统计学意义;结论：枸杞多糖对家兔AS有治疗作用,且注射比口服效果好;枸杞多糖能够通过影响粥样斑块处α-SMA和TGF-β的表达,稳定斑块进而使AS斑块消退,对AS起到治疗作用。

THE STUDY OF THERAPEUTICAL EFFECT OF LYCIUM BARBARUM POLYSACC HORIDES ON ATHEROSCLEVOSIS

Objective：In the present study,we provided AS model animal,with LBP detected the expression of α-SMA,TGF-β in the AS plaque tissue and observed morphology for AS plaque tissue,and to approach further LBP therapeutical action and mechanism of action in AS.Methods： IMale healthy New Zealand white rabbits were randomly divided into 4 groups of eight animals： normal control group,model control group,orally therapic group,injection therapic group.The normal control group animals were always provided with standard feedstuff,the ethev groups animals received hypsi-cholesterol feedstuff for 10 weeks.In the next 10 weeks,model control group animals changed to receive standard feedstuff,orally therapic group animals received standard feedstuff＋LBP（20 mg/kg,taking orally）,injection therapic group animals received standard feedstuff＋LBP（3 mg/kg,intraperitoneal injection）.At the end of 20 weeks,all rabbits were killed to perform the pathologic examinations.In addition,the expressions of α-SMA and TGF-β in the AS plaque tissue were analyzed by Masson dyes and immunohistochemistry.Results： Tthe therapic groups were obvious better as compared with the model control group.The positive expression of these proteins in the injection group rabbits were more noticeable than those observed in the other group rabbits.Conclusions： ILBP has therapeutic action on the rabbit AS,and injection effective is better than those of oral administration;LBP could influence the expressions of α-SMA and TGF-β in the AS plaque tissue,stable plaques and resolve AS plaques,to treat further AS through impacting the plaque quality.
