可生物降解的聚乳酸阿霉素对胶质瘤缓释化疗系统的研制及实验研究

摘    要:目的研制一种可生物降解的肿瘤间质内缓释化疗系统,提高和维持胶质瘤内药物浓度,降低全身毒副作用。方法应用聚乳酸和盐酸阿霉素制成聚乳酸-阿霉素缓释片,经高效液相色谱仪测定其缓释速率,动物实验检测其生物相容性、体内杀瘤效果和全身毒副作用。结果 该缓释系统7天可将C6胶质瘤动物模型皮下肿瘤全部杀灭,骨髓抑制较同剂量静脉给药减少50%,且程度较轻,无死亡率,无胃肠道反应,脱毛现象减少60%,对正常组织细胞无刺激损害。结论聚乳酸-阿霉素缓释系统杀瘤效果肯定,毒副作用少,具有良好的生物相容性。

Abstract:Objective TO develop a biodegradable drug controlled-release system to improve theconcentration of chemotherapeutant in brain glioma and reduce its toxicity. Methods Adriamycin-PLA was made by mixingthem in a certain proportion. Its releasing rate was detected with HPLC. The biocompatibility, cell toxicity,chemotherapeutic effect and side effects was tested in rat glioma model. Results The C6 glioma in rats model was eliminated in 7 days with slight bone marrow depression. The Adriamycin-PLA has not caused inflammatory reaction.Conclusions Adr-PLA controlled-releasing system has a good killing effection ac glioma in rat, good biocompatibility, with slight side effects.
