烟曲霉感染对人脐静脉内皮细胞凋亡的影响

摘    要:目的观察烟曲霉菌丝感染后人脐静脉内皮细胞(HUVEC)的凋亡情况。方法建立烟曲霉菌丝感染HUVEC的体外模型。实验分3组:①空白对照组,细胞不予任何刺激;②烟曲霉菌丝组Ⅰ(浓度10~5CFU/mL);③烟曲霉菌丝组Ⅱ(浓度10~6 CFU/mL)。流式细胞术分别于2、4和8 h 3个时相点检测HUVEC凋亡率。结果与2 h比较,暴露于不同浓度烟曲霉菌丝的HUVEC凋亡率在4和8 h逐渐增加(P<0.05);与空白对照组相应时间点比较,烟曲霉菌丝组Ⅰ和烟曲霉菌丝组Ⅱ的HUVEC凋亡率均明显增加(P<0.01),且烟曲霉菌丝组Ⅱ高于烟曲霉菌丝组Ⅰ(P<0.05)。结论烟曲霉菌丝可诱导HUVEC凋亡;一定时间内,随HUVEC接触菌丝时间的延长,凋亡率增加;相同时间点,烟曲霉菌丝浓度越高,HUVEC凋亡率越高。

Abstract:Objective To observe the apoptosis of human umbilical vein endothelial cells (HUVECs) following infection with Aspergillus fumigatus hyphae. Methods Three groups were included in this experiment after the establishment of an in vitro model of t-IUVECs infected with Aspergillus fumigatus hyphae: (1) negative control group., no stimulus to HUVECs; (2)Aspergillus fumigatus hyphae group I: induced by 10^6 CFU/mL of Aspergillus fumigatus hyphae; (3)Aspergillus fumlgatus hyphae group Ⅱ induced by 10^6CFU/mL of Aspergillus fumigatus hyphae. Flow cytometry was used to detect the apoptosis of HUVECs at 2, 4, and 8 h following infection. Results The apoptosis rate of HUVECs increased gradually at 2, 4 and 8 h in Aspergillus fumigatus hyphae group I and Ⅱ (P〈0.05). Compared to negative control group at the corresponding time points, the apoptosis rate of HUVECs in Aspergillus fumigatus hyphae groups increased significantly at 2, 4, and 8 h (P〈 0.01 ). Moreover, it was higher in Aspergillus fumigatus hypha group Ⅱ than that in group I (P〈0.05). Conclusions Aspergillus fumigatus hyphae can induce HUVECs apoptosis. Within certain period of time, apoptosis rate increases with the time of hyphae induction of HUVECs. At the same time point, the higher the concentration of Aspergillus fumigatus hyphae, the higher apoptosis rate of HUVECs.
