神经干细胞脑内移植对血管性痴呆大鼠认知功能的治疗研究

摘    要:目的研究神经干细胞脑内移植对双侧颈总动脉永久性结扎(2VO)所致血管性痴呆(VD)大鼠认知功能的改善作用。方法以2VO法来制备大鼠VD模型,用Morris水迷宫实验来检测各组大鼠学习记忆能力的差异;取VD组大鼠给予神经干细胞脑内移植,观察移植后VD大鼠认知功能的改善情况。结果神经干细胞脑内移植后,可在大鼠脑内分化为神经元,Morris水迷宫测试表明,VD治疗组大鼠的潜伏期缩短,平均速度明显加快。结论神经干细胞脑内移植后可以改善由VD引起的大鼠认知功能的减退。

Abstract:Objective To study improvement of cognitive function impairment of vascular dementia rats induced by a permanent bilateral ligation of common carotid arteries(2VO) after transplantation of neural stem cell in brain.Methods VD model of rats was established by 2VO.The Morris water test was performed to detect difference in the cognitive function between VD group and control group.After transplantation of neural stem cell in brain,cognitive function improvement of VD treatment group was observed.Results Neural stem cell could be differentiated into mature neuron after transplantation into rat brain.Morris water test showed that latency time was shorter and average velocity was increased in VD treatment group.Conclusion Cognitive function impairment could be improved by transplantation of neural stem cell in brain in VD rats.
