多糖对人胃癌SGC7901细胞生长抑制的研究

摘    要:目的研究多糖对人胃癌SGC7901细胞生长的抑制作用。方法培养人胃癌SGC7901细胞,采用多糖处理后观察其形态变化、凋亡率、BCL-2蛋白及BAX蛋白的表达。结果多糖可以抑制入胃癌SCC7901细胞的增殖,并呈现出对剂量和时间的依赖性。用10mg/ml的多糖处理培养细胞48h后,细胞凋亡率达到39.72%;并出现明显的DNA片段化;BCL-2蛋白的表达下降,而BAX蛋白的表达增加。结论多糖可以通过诱导细胞发生凋亡来抑制入胃癌SGC7901细胞的生长,从而达到}台疗胃癌的目的。

Abstract:In this study, transformations of the cultured human gastric cancer cells (SGC7901)treating with polysaecharide were investigated. The results showed that multiplication of the cultured cells was restrained by treating with polysaccharide, and more cells' multiplication was restrained because of increasing in the polysaccharide dosage and treating time. The cultured cells performed typical apoptosis both in morphological characters and DNA fragmenta- tion after treating 48 hours with 10mg/ml polysaccharide. There were about 40% of the cultured cells performed apop- tosis, and expression of BCL - 2 protein was down - regulated and BAX protein was up - regulated. So, treating with polysaccharide can induce apoptosis and restrain cell multiplication, which is used for human gastric cancer therapy.
