Abstract:Objective: To investigate the cytotoxicity of cytokine-induced killer (CIK) cells against human esophagus carcinoma cell line(EC9706).Methods: Expression of NKG2D ligands on EC9706 cells were analyzed by flow cytometry.Cytotoxicity of CIK cells against EC9706 cells was measured using a standard LDH releasing assay.Effect of CIK cells on cancer was tested by nude mice in vivo.Results: It was found that MICA,ULBP2 were expressed,MICB,ULBP1,ULBP3 were not detectable by EC9706 cells.At 10∶1,20∶1 and 30∶1 E∶T ratios,cytotoxicity of CIK cells against cancer cells were respectively(28.81±0.47)%,(37.78±0.22)% and(44.31±1.06)%(P<0.05),and these results indicated that cytotoxicity of CIK cells against cancer cells gradually increased with E:T ratios increasing.Correlation Analysis indicated that the activity of CIK cells against EC9706 cells was closely correlated to the NKG2D and CD3~+CD56~+ expressions.At 30:1 E:T ratio,the cytolytic activity of the CIK cells at before and after NKG2D Antibody closure were respectively(44.31±1.06)% and(30.29±1.45)%(P<0.05).The differences in times of the tumor forming and tumor weight between control group and treatment group were statistically significant(P<0.01),and tumor inhibitory rate of treatment group was 35.19%.In addition,histopathology showed that there were more lymphocytes infiltration and necrosis regions in tumor tissue in the treatment group than that in the control group. Conclusion: Our data suggested that CIK cells may be used as a novel adoptive immunotherapy for patients with esophagus carcinoma.
