摘    要:目的通过研究苦参素对裸鼠胃癌移植瘤组织中结合免疫球蛋白(BIP)表达的影响,以探讨其诱导肿瘤凋亡的可能机制。方法运用移植瘤体积判定肿瘤生长抑制,TUNEL检测细胞凋亡率,原位杂交技术显现BIP表达状况。结果经不同浓度苦参素处理9 d后,肿瘤平均体积出现明显缩小(P0.05);各剂量组均出现细胞凋亡(P0.01);BIP表达明显下降(P0.01)。结论苦参素诱导裸鼠移植瘤凋亡的作用与调控内质网应激信号通路相关,BIP基因下调可能是其作用靶点之一。

Impacts of Marine on BIP Expression in Transplanted Tumor Tissue of Gastric Cancer in Nude Mice

Abstract:Objective To study the impacts of marine on BIP expression in transplanted tumor tis- sue of gastric cancer in nude mice so as to explore the potential mechanism on tumor apoptosis. Methods The transplanted tumor volume was used to determine the suppression of tumor growth. TUNEL was adopted to detect apoptosis rate. In situ hybridization (1SH)was applied to indicate BIP expression. Results After treatment of marine at different concentrations for 9 days, the average tumor volume was getting smaller appar- ently(P 〈0.05). The apoptosis presented in every dose group(P 〈0. 01 )and BIP expression was reduced obviously ( P 〈0. 01 ). Conclusion The inductive effect of marine on the apoptosis of transplanted tumor in nude mice is related to the regulation of endoplasmic reticulum stress signal channel. The down - regulation of BIP gene could be one of its effect targets.
