CA11对胃癌细胞SGC-70901的放疗增敏作用

目的研究CA11基因对人胃癌细胞株SGC7901放疗增敏的作用。方法以pcDNA3.1-CA11质粒转染人胃癌细胞株SGC7901后对细胞株行放射处理,Western blot、RT-PCR检测凋亡相关酶Caspase-3的表达,MTT法检测细胞增殖情况,流式细胞术检测细胞凋亡情况。结果 pcDNA3.1-CA11质粒转染的SGC7901放疗72 h后Caspase-3在mRNA、蛋白水平表达明显上调,CA11转染组细胞增殖抑制率为(35.41±3.78)%,较对照组的(7.32±0.92)%、空质粒转染组的(10.06±1.47)%,明显升高(均P0.05);CA11转染组细胞凋亡率为(42.19±7.17)%,较对照组的(7.79±1.01)%、空质粒转染组的(20.43±6.24)%,明显升高(均P0.05)。结论 CA11基因对胃癌细胞株SGC7901细胞具有放疗增敏作用。

The role of CA11 in radiotherapy sensitization of human gastric cells SGC7901

Objective To explore the radiotherapy sensitization mechanism of CA11 gene in human gastric cancer cells SGC7901. Methods People gastric carcinoma cell line SGC7901 transfected by pcDNA3. 1-CA11plasmid was followed by radiation treatment. Western blot,RT-PCR detected the activity of apoptosis related enzyme Caspase-3. MTT detected cell proliferation,and flow cytometry detected cell apoptosis. Results The mRNA and protein expression level of Caspase-3 raised obviously after 72 h of radiotherapy in pcDNA3. 1-CA11plasmid transfection cells. The cellular proliferation inhibition rate of CA11 transfection（ 35. 41 ± 3. 78） % was significantly increased compared with the control group（ P 0. 05）; the apoptosis rate of CA11 transfection（ 42. 19 ± 7. 17） % was significantly increased compared with the control group（ P 0. 05）. Conclusions CA11 possess radiotherapy sensitization effect with SGC7901.
