摘    要:目的:研究高血脂对主动脉血管细胞粘附分子1(VCAM-1)基因表达的影响。方法:采用胆固醇喂饲法建立兔高血脂模型,在喂饲后1,6周处死动物,应用组织原位杂交、免疫组织化学等方法检测产动脉VCAM-1的表达,并与正常组对比。结果:喂饲胆固醇1周后高血脂很快诱导主动脉内皮细胞表达VCAM-1,并在6周后动脉粥样硬化(AS)病灶内也有丰富表达。结论:高血脂促进动脉VCAM-1表达,是其促进AS发病的机制

Abstract:Objective: Circulating monocyte binding to vascular endothelial cell is an important step in atherosclerosis. Vascular cell adhesion molecule 1(VCAM 1) has a pivotal role in mediating adhesion of these two kinds of cells. The current study was designed to evaluate the effect of hypercholesterolemia on inducing gene expression of VCAM 1 in aortas of rabbits. Methods: Rabbits were fed with cholesterol to create hypercholesterolemic animal model. At the end of 1st week and 6th week, the animal were killed. Using in situ hybridization and immunohistochemistry, we check the expression of VCAM 1 in aorta. Results: we found that hypercholesterolemia quickly induced gene expression of VCAM 1 in the aortic endothelial cells after 1 week of cholestrol feeding, and there was also a high expression in the atherosclerosis lesion 5 weeks later. Conclusion: These results indicate that inducing gene expression of VCAM 1 is one of atherogenic mechanisms of hypercholesterolemia.
