1-甲基乙内酰脲与氟尿嘧啶联合对人结肠癌SW480细胞增殖和凋亡作用的研究

目的探讨1-甲基乙内酰脲(1-methylhydantoin)单用以及与氟尿嘧啶(5-Fu)联合对结肠癌SW480细胞增殖抑制作用和诱导凋亡。方法使用MTT比色法检测1-甲基乙内酰脲M_((a-h))在不同浓度(0.5、5、10、20、50、100、200、500 mg/L)单用及与不同浓度5-Fu_((a-d))(10、20、50、100 mg/L)联合作用于人结肠癌SW480细胞的吸光度值,然后再计算抑制率(IR)。流式细胞仪监测M_d、M_e、Fu_b、M_d+Fu_b、M_e+Fu_b处理SW480细胞48 h对细胞周期分布和凋亡影响。结果 1-甲基乙内酰脲在体外对人结肠癌SW480细胞有增殖抑制作用。在低浓度时,1-甲基乙内酰脲对人结肠癌SW480细胞增殖抑制效果与5-Fu相当(P0.05);在高浓度时,1-甲基乙内酰脲对人结肠癌SW480细胞抑制增殖效果弱于5-Fu(P0.05)。1-甲基乙内酰脲与氟尿嘧啶联合应用有协同抗肿瘤效果。结论 1-甲基乙内酰脲能够抑制人结肠癌细胞SW480的增殖和诱导凋亡作用,并与氟尿嘧啶有协同抗瘤细胞增殖效果。

Study on the effects of 1-methylhydantoin combined with fluorouracil on the proliferation and apoptosis of human colon cancer SW480 cells

Objective To investigate the proliferation inhibitory effect and inducingapoptosis of 1-methylhydantoin alone and combined application with 5-fluorouracil(5-Fu)on human colon cancer SW480 cells.Methods MTT colorimetric assay was used to detect the absorbance value of colon cancer SW480 cells after the application of different concentrations of 1-methylhydantoin(0.5,5,10,20,50,100,200,500 mg/L)alone and combined application with 5-Fu (10,20,50,100 mg/L),and then the inhibitory rate was calculated.Flow cytometry was used to monitor the cell cycle distribution and apoptosis of SW480 cells treated with Md,Me,Fub,Md+Fuband Me+Fubfor 48 h.Results 1-methylhydantoin had an inhibitory effect on the proliferation of human colon cancer SW480 cells in vitro.At low concentration, the inhibitory effect of 1-methylhydantoin on the proliferation of human colon cancer SW480 cells was similar to that of 5-Fu(P>0.05);at high concentration,the inhibitory effect of 1-methylhydantoin on the proliferation of human colon cancer SW480 cells was weaker than that of 5-Fu(P<0.05).Combined application of 1-methylhydantoin and 5-Fu had a synergistic anti-tumor effect.Conclusion 1-methylhydantoin can inhibit the proliferation and inducingapoptosis of human colon cancer cell SW480 and has a synergistic effect with 5-Fu.
