RUNX2基因rs1406846位点多态性与中国汉族女性TMJOA的相关性研究

摘    要:目的本研究旨在评价RUNX2基因rs1406846位点多态性与中国汉族女性颞下颌关节骨关节炎的相关性。方法依据颞下颌关节紊乱病研究诊断标准(RDC/TMD),共纳入240例受试者,包括114例TMJOA患者和126例健康的女性对照。运用测序的方法检测RUNX2基因上rs1406846位点的多态性,并采用Logistic回归模型对基因型、等位基因、携带频率分布进行统计学分析(SPSS 20.0)。结果中国汉族女性TMJOA组中携带RUNX2-rs1406846上TT基因型和对照组的AA基因型相比具有统计学差异(P=0.032),T等位基因频率在TMJOA组中明显高于健康对照组(64.0% vs 54.4%,P=0.032)。结论单核苷酸多态性研究显示,在中国汉族女性的TMJOA人群中RUNX2基因和TMJOA具有相关性,其中T等位基因可能是该疾病的危险因素。

Study on the correlation between rs1406846 polymorphism of RUNX2 gene and TMJOA in Chinese Han females
Abstract:Objective To evaluate the association between rs1406846 polymorphism of RUNX2 gene and temporomandibular joint osteoarthritis in Chinese Han females.Methods A total of 240 subjects,including 114 TMJOA patients and 126 healthy female controls were enrolled,according to the diagnostic criteria for temporomandibular disorders(RDC/TMD).The polymorphism of rs1406846 locus of RUNX2 gene was detected by sequencing.And the genotype, allele and frequency distribution were analyzed by Logistic regression model(SPSS 20.0).Results There was statistically significant difference between TT genotype of RUNX2-rs1406846 gene in the Chinese Han female TMJOA group and AA genotype in the control group(P=0.032).T allele frequency was significantly higher in the TMJOA group than that in the healthy control group(64.0%vs 54.4%,P=0.032).Conclusion The single nucleotide polymorphism study shows that there is correlation of RUNX2 gene and TMJOA in the TMJOA population of Han Chinese women,and the T allele might be the risk factor of the disease.
