摘    要:目的:探讨N-乙酰半胱氨酸(NAC)对软脂酸培养下胰岛细胞的影响。方法:体外培养胰岛细胞INS-1,分为空白对照组,软脂酸组(0.25 mmol/L),NAC作用组(浓度分别为50μmol/L,100μmol/L,200μmol/L),培养24h后,取上清测定胰岛素水平及GSH-Px、MDA含量。结果:软脂酸可使胰岛素分泌量减少,GSH-Px浓度下降,MDA含量增加,NAC可以明显抑制软脂酸引起的上述变化,抑制作用随NAC浓度的增加而增强。结论:抗氧化剂NAC对软脂酸导致的胰岛细胞损伤有一定的保护作用,并且呈浓度依赖性。

Effect of N-acetylcysteine on islet cell cutueled with palmitic acid

Abstract:Objective:To investigate the effect of N-acetylcysteine on islet cell cultrued with palmitic acid.Methods:The cultrued islet cells(INS-1)were divided into 5 groups:normal control group,palmitic acid(PA) group(0.25mmol/L),50μmol/L NAC+ PA group,100μmol/L NAC+ PA group,200μmol/L NAC+ PA group.The level of insulin,GSH-Px and MDA in Culture supernatant were measured when cultrued for 24h.Result:Palmitic acid had the effects of inhibiting insulin secretion of pancreatic islet cell,reducing the concentration of GSH-Px and enhancing the level of MDA.This change were inhibited by NAC with a dose-dependent manner.Conclusion:NAC protected islet cell against injury induced by palmitic acid through inhibiting oxidation in dose-dependent manner.
