摘    要:目的:探讨Hedgehog信号通路特异性阻滞剂GANT61抑制本通路在胰腺癌细胞株中的表达,观察胰腺癌细胞株增殖、迁移等能力的改变,为胰腺癌的治疗提供基础实验数据。方法:采用多株胰腺癌细胞,半定量RT-PCR法从中检测出Hedgehog通路表达相对活跃的细胞株,采用不同浓度的GANT61处理细胞株后,MTT法检测细胞株增殖活力改变,细胞划痕实验检测细胞迁移能力改变。结果:采用GANT61处理后,随着浓度的增加,胰腺癌细胞株增殖活力下降,迁移能力降低。结论:GANT61可通过抑制Hedgehog信号通路的表达,有效抑制胰腺癌细胞株的增殖、迁移等,为GNAT61药物在临床治疗胰腺癌提供扎实的理论基础。

Abstract:Objective:To explore the effects of GANT61inhibiting Hedgehog signaling pathway on proliferation and migration of human pancreatic cancer cells.Methods:Selected one cell line which has the relatively high activation in Hedgehog signaling pathway from four pancreatic tumor cell lines by semiquantity RT-PCR,detected cell proliferation viability by MTT and cell migration ability by cell scarification experiment after treatment of GANT61at different concentrations.Results:With the increase of GANT61concentration,the proliferation activity was decreased and migration ability was reduced in the pancreatic tumor cell.Conclusion:GANT61can effectively inhibit the proliferation and migration of pancreatic cancer cell lines by restricting Hedgehog signaling pathway.
