银杏内酯B、C及联合用药预处理对大鼠心肌缺血/再灌注心功能的影响

摘    要:目的 探讨银杏内酯B(ginkgolide B,GB)、银杏内酯C(ginkgolide C,GC)及两者1∶1联合用药预处理对心肌缺血/再灌注损伤(ischemia/reperfusion injury,I/R)大鼠血流动力学的影响及其与氧自由基的关系.方法 50 只SD大鼠,随机分为5 组(n=10):正常对照组(Control组)、缺血/再灌注组(I/R组)、银杏内酯B预处理组(GB组)、银杏内酯C预处理组(GC组)及银杏内酯B和C联合用药预处理组(GB+GC组).采用Langendorff装置逆行插管灌流,建立大鼠离体心脏I /R 模型;除Control组外,其余各组均缺血20 min,再灌注40 min.记录左心室收缩压(LVSP)、左心室舒张末压(LVEDP)、左心室内压上升和下降的最大变化速率(+dp/dtmax),并测定冠状动脉流出液中乳酸脱氢酶(LDH)的活性.结果 GB组、GC组及GB+GC组用药预处理后,复灌时左心室内压差(△LVP)、+ dp/dtmax等心功能指标明显优于I/R组(P〈0.05),联合用药较单一用药效果差异不明显,各用药组的LDH活性均较I/R组明显降低.结论 银杏内酯B、C及联合用药均能一定程度改善大鼠心肌缺血/再灌注损伤,但联合用药较单一用药效果优势不明显.

Abstract:Objective To explore the effects of ginkgolide B, ginkgolide C and their combined therapy on the cardi- ac function of rats with ischemia/reperfusion injury. Methods Fifty Sprague - Dawley rats were randomly divided into five groups: a normal control group (Control group), an ischemia/reperfusion injury group (I/R group), a ginkgolide B pretreatment group (GB group), a ginkgolide C pretreatment group (GC group) and a combined pretreatment group (GB + GC group). Langendorff apparatus was adopted to perform retrograde perfusion, and an in vitro isolated rat heart model of I/R injury was established. Each group except the control underwent ischemia operation for 20 minutes followed by reperfusion for 40 minutes. Then, we recorded left ventricular systolic pressure ( LVSP), left ventricular end diastolic pressure ( LVEDP), the maximal rate of systolic pressure rise ( + dp/dtmax ) and the maximal rate of diastolic' pressure decrease ( - dp/dtm,x ) and detected the activity of LDH in the fluid of coronary artery. Results During reperfusion, the GB group, GC group and GB + GC group presented remarkable increases in LVP and decreases in the activity of LDH ( P 〈 0.05 ), There was no significant change between the effects of lhe (;B + GC group and GB or GC group, Conclusion GB, GC and their combination can improve the cardiac fu.ction of rats with I/R injury, but the combined pretreatment did not present advantages over GB or GC alone.
