当归内酯类成分的结构鉴定与抗氧化活性筛选

摘    要:目的分离当归中内酯类成分,并对其血管内皮细胞保护活性进行筛选。方法当归经CO2超临界萃取,萃取物通过硅胶柱层析分离得到单体成分,波谱法鉴定其结构;采用过氧化氢(H2O2)建立体外培养的人脐静脉内皮细胞(HU-VEC)氧化应激损伤模型,设立模型组、对照组和药物组,其中药物组给予5种当归内酯类成分预培养24 h后加入25mmol/LH2O2,四甲基偶氮唑盐(MTT)法检测细胞存活率。结果分离得到(Z)-藁本内酯(Ⅰ),(Z)-正丁烯基苯酞(Ⅱ),exo-Z,Z’-3α.7α’,7α.3α’-双藁本内酯(Ⅲ),3,9-二羟基藁本内酯(Ⅳ),(Z,Z’)-6.8’,7.3’-双藁本内酯(Ⅴ)等5种内酯类成分。其中(Z)-藁本内酯、3,9-二羟基藁本内酯具有显著的血管内皮细胞保护活性。结论当归中某些内酯类成分具有较强的抗氧化能力及内皮细胞保护作用,符合当归的功效、主治。
