摘 要: | 目的研究白藜芦醇(RES)对60Coγ射线照射小鼠骨髓细胞凋亡的影响。方法30只雄性ICR小鼠随机分成5组,即正常对照组、照射对照组以及50mg/kg照射组、100mg/kg照射组和RES200mg/kg照射组,照射前3d灌服给药,每天1次。除正常对照组外,所有小鼠60c01射线全身5.5Gy辐照1次,照射后6h取小鼠骨髓细胞,采用单细胞凝胶电泳实验检测骨髓细胞DNA损伤,流式细胞术检测骨髓细胞凋亡率和CD95的表达。结果与正常对照组比较,照射对照组小鼠骨髓细胞的尾长、Olive尾矩、尾分布矩和尾DNA百分含量显著增加,P〈0.05,骨髓细胞凋亡率显著增加,P〈0.05,CD95表达百分率显著增加,P〈0.05;与照射对照组比较,RES各给药组小鼠骨髓细胞的尾长、Olive尾矩、尾分布矩和尾DNA百分含量显著降低,P〈0.05,骨髓细胞晚期凋亡率的显著降低,P〈0.05,RES中、高剂量组的CD95表达百分率显著降低,P〈0.05。结论RES可以减少60Coy射线照射引起的小鼠骨髓细胞凋亡,可能与抑制了辐射引起的CD95的高表达有关。

Abstract: | Objective To investigate effects of resveratrol on bone marrow cell apoptosis in 60 Co-gamma rays irradiated mice. Methods 30 male ICR mice were randomized into 5 groups, including control group, irradiated group, HESS0 mg/kg irradiated group, RES100 mg/kg irradiated group and RES200 mg/kg irradiated group. Resveratrol was given for three days prior to irradiation. Except for the control group, all mice were whole-body irradiated by 60 Co gamma ray with a single dose of 5.5 Gy. After being executed, the mice' s bone marrow cell were measured 6 hours after irradiation. DNA damage of the cell was detected by single cell gel electrophoresis (SCGE) assay. Apoptosis rate and expression of CD95 were detected by flow cytometry. Results Compared with the control group, the tail length, olive tail moment, tail moment and DNA percentage of bone marrow Cell of the irradiated group were significantly increased (P 〈0.05), as well as apoptosis percentage and expression percentage of CD95 (P 〈0.05); Compared with the irradiated group, tail length, olive tail moment, tail moment and tail DNA percentage of bone marrow cells, late apoptosis percentage of Resveratrol each treatment group was significantly lower (P 〈0.05), and CD95 expression rate in the percentage of high-dose group was significantly lower (P 〈0.05) in Resveratrol 100 mg/kg and 200 mg/kg treatment group. Conclusion Resveratrol is considered being effective in reducing the bone marrow cell apoptosis induced by 60Co γ irradiation in mice, which may be related to inhibiting the high expression of CD95 induced by irradiation. |
