V79和V798细胞周期及其调控因子的差异比较

为了解Ｖ７９和Ｖ７９－８两种细胞及调控因子的差异，用「＾３Ｈ『胸腺嘧核苷参入法检测两种细胞，发现Ｖ７９细胞无可检测到的Ｇ２期；Ｖ７９－８细胞则同时缺管可测到的Ｇ１和Ｇ２期。流式细胞光度术显示，Ｖ７９Ｇ１ＡＤＷＥ调控上存在差异。比较两者Ｇ１期周期调控因子的ｍＲＮＡ表达水平发现，Ｖ７９－８ｃｙｃｌｉｎＤ１，ｃｙｃｌｉｎＤ３和ＩＤ表达水平明显高于Ｖ７９，这可能直接导致了两者Ｇ１的差异。

DIFFERENTIAL CELL CYCLE REGULATION BETWEEN V79 AND V79-8 CELLS

V 79 and V 79 8 are two Chinese hamster lung fibroblast cell lines which proliferate very rapidly.It is verified through [ 3 H] thymidine incorporation assay that V 79 cells have non detectable G2 phase and V 79 8 cells have neither G1 nor G2 phase.With flow cytometry,we found that V 79 had obviously more G1 phase cells than V 79 8 ,which indicates that they are different in G1 phase regulation.Comparing the mRNA expression levels of several regulators in G1 phase,we also found that the expression levels of cyclin D1,D3 and Id in V 79 8 cells was obviously higher than that in V 79 cells.This might cause the G1 difference between V 79 and V 79 8 cells directly.
