iNOS在碱烧伤角膜新生血管形成中的表达及意义

摘    要:【目的】探讨碱烧伤后角膜新生血管形成过程中诱导型一氧化氮合成酶 (iNOS)的表达及作用。【方法】采用碱烧伤诱导角膜新生血管模型 ,将 2 6只SD大鼠随机分成三组 :A组为对照组 ,B组为治疗组 ,C组为正常组。治疗组给予 1%L NAME滴眼液局部应用。采用裂隙灯照相 ,免疫组织化学染色 ,多媒体彩色图像病理分析系统分别计算d7角膜新生血管面积和iNOS免疫组化染色的平均光密度值。【结果】正常大鼠角膜上皮基底膜和基质层即表达iNOS ,新生血管对照组iNOS表达明显增强 ,L NAME治疗组其表达明显受抑制。【结论】iNOS表达水平与炎性角膜新生血管明显相关 ,局部应用 1%的L NAME滴眼液通过抑制iNOS的表达而产生抗新生血管的作用。

Abstract:ObjectiveTo investigate the expression of inducible nitric oxide synthase(iNOS) in cornea after alkaline burn.Corneal neovascularization(CNV) model was established by putting the filter paper with the size of 3mm in diameter immersed in 1 mol/L NaOH solution on the rat cornea for 90 seconds. Twenty six Sprague-Dawley rats were randomly divided into 3 groups : group A as control group, group B as treated one with topically applied 10 g/L NG-Nitro-L-Arginine Methyl Ester (L-NAME) drops and group C as normal one. Slit-lamp biomicroscopy and photography, immunohistochemical staining were done; The area of CNV were calculated and the average optic density of immunohistochemical staining of iNOS were measured by imaging analysis system.The normal rat corneas expressed iNOS in the basis of epithelium and stroma. The expression of iNOS increased markedly in group A and were suppressed significantly in group B applied topically with 10 g/L L-NAME drops.Conclusion]The levels of iNOS were markedly related to inflammatory corneal neovascularization and topical application of 10 g/L. L-NAME are effective in the inhibition of corneal neovascularization by inhibiting the iNOS expression in rat cornea after alkaline burn.
