丹参酮加强电针对去卵巢大鼠下丘脑-垂体-卵巢轴异常功能的调整作用

摘    要:目的 通过观察去卵巢大鼠外周血雌激素 (E2 )水平 ,肝脏和脂肪组织芳香化酶 (雌激素合成关键酶 )表达的变化 ,探讨丹参酮对去卵巢大鼠下丘脑 -垂体 -卵巢轴 (HPOA)异常功能的影响 ;及丹参酮能否加强针刺调整 HPOA异常功能的作用。方法 动物分成正常组 (INT)、去卵巢组 (OVX)、去卵巢 丹参酮组 (OVX T)、去卵巢 电针组 (OVX EA )和去卵巢 电针 丹参酮组 (OVX EA T) ;采用 Western- blot和放射免疫分析法 ,测定肝脏和脂肪组织芳香化酶蛋白表达及血 E2 的水平。结果  OVX T和 OVX EA大鼠脂肪组织和肝脏芳香化酶蛋白表达和血 E2 水平都比 OVX明显增加 (P<0 .0 5 ) ;OVX EA T大鼠脂肪组织芳香化酶蛋白表达和血 E2 水平 ,比 OVX EA显著升高 (P<0 .0 5 ) ,但肝脏组织的酶蛋白表达无明显改变。结论 丹参酮可能通过促进去卵巢大鼠脂肪和肝组织芳香化酶蛋白表达 ,对去卵巢大鼠 HPOA功能异常有一定的治疗作用 ;丹参酮可能有加强电针调整 HPOA异常功能的作用。

Abstract:Objective To investigate in ovariectomized rats the influence of Tanshinone on the dysfunction of hypothalamic-pituitary-ovarian axis (HPOA) and if Tanshinone strengthens the regulative effect of electroacupuncture on HPOA dysfunction by observing changes in blood estrogen (E 2) level and the expression of hepatic and adipose tissue aromatase (key estrigen synthetase).Methods The animals were randomly divided into normal (INT), ovariectomy (OVX), ovariectomy Tanshinone (OVX T), ovariectomy electroacupuncture (OVX EA) and ovariectomy electroacupuncture Tanshinone (OVX EA T) groups. The expression of hepatic and adipose tissue aromatase and blood E 2 level were measured by Western-blot and radioimmunoassay.Results The expression of adipose and hepatic tissue aromatase and blood E 2 level were significantly increased in OVX T group and OVX EA group as compared with OVX group (P< 0.05). The expression of adipose tissue aromatase and blood E 2 level were significantly higher in OVX EA T group than in OVX EA group (P< 0.05), but there was no significant change in hepatic tissue aromatase.Conclusion Tanshinone may produce a certain curative effect on ovariectomized rat's HPOA dysfunction by promoting the expression of adipose and hepatic tissue aromatase. Tanshinone may strengthen the regulative effect of electroacupuncture on HPOA dysfunction.
