缺氧缺血性脑损伤新生大鼠IL-16的表达及意义

摘    要:目的观察缺氧缺血性脑损伤(HIBD)新生大鼠白细胞介素-16(IL-16)表达的变化。方法建立新生大鼠缺氧缺血(HI)模型,实验分为假手术组、HIBD组。采用酶联免疫吸附法(ELISA)检测假手术组手术后和HIBD组缺氧缺血后3h、12h、24h、3d、7d、14d、28d脑组织匀浆中IL-16的水平。结果IL-16在HIBD组3h开始上升,24h达高峰,3d开始下降,28d基本恢复正常。与假手术组相比,HIBD组的IL-16水平在缺氧缺血后3h、12h、24h、3d、7d、14d均明显升高(P〈0.01),而28d无明显差异(P〉0.05)。结论HIBD新生大鼠IL-16蛋白表达的增加出现早、持续时间长,表明IL-16参与HIBD的发病过程。

Abstract:Objective To observe the variations in the expression levels of interleukin - 16 (IL - 16) protein in neonatal rats with hypoxie - ischemic brain damage (HIBD). Methods Healthy 7 - day - old Sparague - Dawley (SD) rats were randomized into two groups: sham group and HIBD group. The two groups were subdivided into groups according to the time points : 3 h, 12 h, 24 h, 3 d, 7 d, 14 d and 28 d after HI insult. Level of IL - 16 in brain homogenates of HIBD was determined by ELISA. Results Compared with sham group, the increasing expression of IL - 16 in left brain homogenates in HIBD group was observed 3 h after HI insult, peaked at 24 h, then gradually declined at 7 d, and restored to normal at 28 d. The expression of IL - 16 in HIBD group at 3 h, 12 h, 24 h, 3 d, 7 d; 14 d was significantly higher than that of sham group after HI insult ( P 〈 0.01 ) , while the level of IL - 16 in HIBD group had no obvious difference compared with sham group at 28 d ( P 〉 0.05 ). Conclusion The early increase and sustained duration of IL - 16 protein expression level in neonatal rats with HIBD indicated that IL - 16 was involved in HIBD.
