Correlation Analysis Between the Efficacy of Platinum-based Non-small Cell LungCancer and the MTHFR C677T Gene Polymorphism

Objective To investigate the efficacy of platinum-based treatment of non-small cell lung cancer and its correlation with the polymorphism of methylenetetrahydrofolate reductase(MTHFR)C677T gene.Methods 180 patients with NSCLC who underwent a platinum-based chemotherapy regimen after pathological diagnosis and surgical resection were selected as the research group,and 100 healthy people who underwent physical examination were the control group.MTHFR C677T genotyping was performed on DNA extracted from hematology specimens.Compare the prognosis of NSCLC patients with different genotypes of MTHFR C677T.The multivariate Cox proportional hazards regression model was used to analyze the factors affecting the overall survival(OS)of NSCLC patients who received platinum-based adjuvant chemotherapy after surgery.Results 180 NSCLC patients underwent MTHFR C677T genotyping,40 cases of CC genotype(22.22%),73 cases of CT genotype(40.56%),and 67 cases(37.22%)of TT genotype.The frequency of MTHFR C677T CC genotype in the study group was higher than that of the control group(P<0.05),and there was no significant difference in the distribution frequency of CT and TT genotypes between the 2 groups(P>0.05).The frequency of the C allele in the study group was significantly higher than that of the control group,and the frequency of the T allele was lower in the control group(P<0.05).The median survival time of MTHFR C677T genotypes CC,CT,and TT genotypes were 20 months,25 months,and 30 months,respectively,and the difference was significant(P<0.05).The results of the Cox risk ratio model analysis showed that the MTHFR C677T genotype CC was an independent factor of OS(HR=1.07,P=0.012).Conclusion MTHFR C677T genotype is correlated with the efficacy of platinum-based NSCLC treatment.Genotype CC is an independent risk factor affecting OS in patients.
