醋酸曲安缩松及丝裂霉素对瘢痕成纤维细胞整联蛋白表达的影响

摘    要:目的:了解醋酸曲安缩松、丝裂霉素对瘢痕成纤维细胞整联蛋白表达的影响,初步探讨两种药物抗瘢痕增生的作用机制。方法:体外培养瘢痕组织成纤维细胞,利用β1,α1~4整联蛋白单克隆抗体,通过原位ELISA技术检测瘢痕成纤维细胞在不同质量浓度醋酸曲安缩松(1,10,50,100μg/ml)和丝裂霉素(1,5,10,20μg/ml)作用下整联蛋白的表达水平。结果:醋酸曲安缩松、丝裂霉素均能不同程度地抑制瘢痕成纤维细胞整联蛋白的表达,抑制效果在实验范围内随药物浓度的增加而提高。其中醋酸曲安缩松对β1,α1~4整联蛋白表达的最大有效抑制浓度(μg/ml)分别为50,100,100,50,100,抑制率分别达16.44%,34.74%,15.92%,37.95%和32.17%;丝裂霉素对它们的最大有效抑制浓度(μg/ml)分别为

Abstract:Objective:To investigate the effect of triamcinolone acetonideand mitomycin on expression levels of β1 and α1-4 integrins in hypertrophic scar-derived fibroblasts,and study the molecular mechanism of above drugs in the treatment of hypertrophic scar.Methods:Hypertrophic scar-derived fibroblasts were cultured in the presence of various concentration triamcinolone acetonide,and mitomycin.The expression levels of β1 and αl-4 integrins in the cell surface were determined by enzyme-linked immunosorbent assay (ELISA).Results:The shape of cultured hypertrophic scar-derived fibroblast was not changed in the presence of various concentration triamcinolone acetonide and mitomycin. The expression of β1 and αl-4 integrins in hypertrophic scar-derived fibroblasts were inhibited to some extent by triamcinolone acetonide and mitomycin.Conclusion:The results suggest that β1 and αl-4 integrins may be an important intermediary factor on triamcinolone acetonide and mitomycin in the treatment of hypertrophic scars.
