摘    要:目的 探讨DNA双链断裂修复基因RAD51的5'-UTR区135G/C多态与我国人群肺癌发病的关联.方法 采用PCR-RFLP技术,检测300对肺癌病例与正常对照RAD51基因135FG/C多态位点的基因型.用SAD9.13软件进行非条件Logistic回归,校正混杂因素的影响,分析该位点变异与肺癌发病的关联.结果 病例及对照组中,皆未检测出135CC基因型.病例组135GC基因型频率显著低于对照组(26.7%比34.7%,P=0.0336).以携带GG野生基因型个体为参照,携带GC基因型能显著减少个体发生肺癌的危险性(调整优势比=0.72;95%置信区间=0.49~0.98;P=0.042).结论 RAD51的5'-UTR区135G/C多态与我国人群肺癌发病存在显著性关联,135C可能是我国人群肺癌发生的一个遗传保护因素.

Abstract:Objective To determine the association between the polymorphism of 135G/C in 5' -UTR region of RAD51 gene and lung cancer in Chinese population. Methods The genotypes of 135G/C in RAD51 gene were identified with PCR-RFLP in 300 lung cancer cases and 300 age- and sex- matched cancerfree controls. SAS 9.13 software was used to analyze the polymorphism and lung cancer through nonconditional logistic regression. Results 135CC genotype neither in cases nor in controls was found. The genotype frequency of 135GC was significantly lower in cases as compared to controls (26.7% vs 34.7%, P=0.0336). Compared with the GG wild type genotype carriers, the carriers of GC heterozygotes were associated with a significantly decreased risk of lung cancer (adjusted 0R=0.72, 95% CI=0.49- 0.98, P=0.042).Conclusion The polymorphism of 135G/C in 5' -UTR region of RAD51 gene is associated with lung cancer in Chinese population, thus the 135C alle may be an inherited protective factor for lung cancer development.
