摘    要:目的:探讨血小板激活因子(PAF)和维甲酸(RA)对多形核中性粒细胞(PMN)功能和肺微血管内皮细胞(PMVEC)ICAM-1和E-选择素(ELAM-1)表达的影响。方法:采用作者创建的无创性分离法分离培养PMVEC;将PAF和RA分别加入培养有PMVEC的96孔板中,观察二者对PMN-PMVEC粘附的影响;ELISA法测定PMVEC上ELAM-1和ICAM-1的表达;测定PAF和RA对PMN的趋化性、聚集和酸性磷酸酶的释放的影响。结果:(l)PMVEC用PAF处理3.5h后,它与PMN的粘附率从57.3%增加到72.8%(P<0.01)。而用PAF处理PMN同样可增加PMN-PMVEC的粘附率。(2)RA可阻断未刺激PMN与PAF刺激的PMVEC之间的粘附,但对PAF刺激的PMN与未刺激的PMVEC之间的粘附却无作用。(3)PAF可增加PMVEC上ICAM-l和ELAM-l的表达、增加酵母多糖激活血清和组胺对PMN的趋化性、增加PMN的聚集和PMN酸性磷酸酶的释放。(4)RA则可抑制PAF诱导的PMN聚集、酸性磷酸酶的释放及PMN对酵母多糖刺激血清和组胺的趋化性,而ICAM-1和ELAM-1的表达却未改变

Abstract:To investigate the effects of platelet-activating factor(PAF) and retinoic acid(RA) on the expression of ELAM-1 and ICAM-1 and the function of neutrophils. Methods: Pulmonary microvascular endothelial cells(PMVECs) were isolated by the method founded by the author himself. PAF and RA were added to PMVECs monolayers grown to confluency on 96 microplates. Then the effects of both on PMN-PMVECs adherence were measured. The expression of ICAM-1 and ELAM-1 on PMVECs were determined by using ELISA. We also determined the changes of PMNs chemotaxis to zymosan-activated serum and histamine, and PMNs aggregation and the release of acid phosphatase from PMNs when treated by PAF and RA. Results: (1)Preincubation of PMNs with PAF for 3. 5h increased the adhesion rate of polymorphonuclear leukocytes ( PMNs) to PMVECs from 57. 3% to 72. 8%(P<0. 01). Preincubation of PMNs with PAF also increased PMN-PMVEC adhesion rate. (2)All-trans retinoic acid blocked the adherence of untreated PMNs to PAF-pretreated PMVECs but not the adherence of PAF-pretreated PMNs to untreated PMVECs. (3) PAF increased the expression of intercellular adhesion molecule-1(ICAM-1) and E-selection(ELAM-1) on PMVECs, PMNs chemotaxis to zymosan-activated serum and histamine, and PMNs aggregation and the release of acid phosphatase from PMNs. (4) Co-incubation of RA inhibited PAF-induced PMN aggregation, the release of acid phosphatase from PMNs, and PMN chemotaxis to zymosan- activated serum and histamine while the expression of ICAM-1 and ELAM-1 did not change. Conclusion: PAF can activate PMNs,increase the expression of the cellular adhesion molecules on PMVECs and promote the adherence of PMN-PMVEC. RA can be used to ameliorate PMN-mediated infiammation.
