大肠粘膜中P~(16)和P~(53)蛋白的表达及意义

目的 :探讨抑癌基因 P16、P53与大肠癌的发生关系及在大肠癌早期诊断中的价值。方法 :采用免疫组化 SABC法检测大肠癌、大肠粘膜异型增生、炎症及正常组织中 P16、P53蛋白的表达。结果 :在大肠癌中 P16 蛋白阳性表达明显低于炎症时及正常组织 ;而 P53蛋白阳性表达明显高于炎症时及正常组织。但两种蛋白阳性表达在大肠癌与大肠粘膜异型增生中无明显差异 ,且在大肠粘膜病变中呈明显负相关。结论 :P16 、P53基因的突变或缺失可能与大肠癌、大肠粘膜异型增生的发生、发展有关 ,检测其表达产物对判断和预测大肠癌的发生有重要价值。

Expression of P15 and P53 protein and their significance in mucosa of large intestine

Objective:To probe into the relation of tumor inhibiting gene P 16 and P 53 with the occurrence of laryer intestine.Methods:Expression of P 16 and P 53 in carcinoma of large intestine,heterologous hyperplasia of mucosa,inflammation and normal tissuess were measured by immunohisto chemical SABC method.Results:Positive expression of P 16 protein in carcinoma of large intestine was significaantly lower than that in inflammatory and normal tissues,but the positive expression of P 53 protein was significantly higher than that in inflammatory and normal tissues.The positive expression of two proteins had no marked difference between carcinoma and heterologous hyperplasia of carge in testine,but had negative correlation in muconspathologic changes of large intesitne.Conclusion:Mutation and defect of P 16 and P 53 gene maybe related with the occrrence and development of carcinoma and heterologous hyperplasia of large intestine and its product of expression has importment value for judgement and prognosis of the occurrance of carcinoma of large intestine.
