宫颈癌FHIT蛋白表达及与临床的相关性

摘    要:目的:探讨候选抑癌基因FHIT产物FHIT蛋白在由正常宫颈上皮过渡至子宫颈癌过程中的表达缺失情况及其临床意义.方法:应用SP免疫组化染色法检测65例子宫颈癌,25例宫颈上皮肉瘤样病变(CIN),14例慢性子宫颈炎,5例正常子宫颈组织中FHIT蛋白的表达情况,并与组织学类型、组织学分级、肌层浸润深度和临床分期等生物学行为进行相关分析.结果:FHIT蛋白在子宫颈癌前病变和宫颈癌组织中的表达明显减少,缺失率分别为56.00%和72.31%,表达缺失率与组织学类型和组织学分级有关(P<0.05),与子宫颈癌的肌层浸润深度和临床分期无关(P>0.05).结论:FHIT表达缺失是子宫颈癌发生的早期分子事件,FHIT蛋白表达缺失是早期诊断子宫颈癌及癌前病变较好的生物学标志.

Abstract:Objective: To study the clinical significance of FHIT expression in transition from normal cervical epithelia to cervical cancers and precancers. Methods: The expression of FHIT was detected by SP immunohistochemical methods on paraffin sections from 65 cervical carcinomas, 25 cervical intraepithelial neoplasia (CIN) samples, 14 chronic cervicitis samples and 5 normal cervix samples. Results: Fhit protein expression was markedly reduced or absent in 47 of 65 (72.31%) cervical carcinomas and 14 of 25 (56%) cervical intraepithelial neoplasia. In cervical carcinoma the loss of FHIT expression was significantly related to the tissue type and the cell grade (P<0.05), but not related to the depth of myometrium invasion and the clinical stage (P>0.05). Conclusions: The loss of FHIT expression is an early event in cervical tumorigenesis. The detection of FHIT protein may be used as a diagnostic biomarker for the earliest cervical precancers.
