肾福康胶囊对慢性肾功能衰竭模型大鼠的抗氧化作用研究

摘    要:目的探讨肾福康胶囊对慢性肾功能衰竭(CRF)模型大鼠的抗氧化作用。方法取健康雄性SD大鼠70只,造模前取尾静脉血,分离血清,进行尿素氮(BUN)含量测定,按BUN水平均衡,随机分为空白组、模型组、阳性对照组及肾福康低、中、高剂量组,每组10只大鼠。模型组、阳性对照组及肾福康低、中、高剂量组采用灌服腺嘌呤致大鼠CRF模型,同时肾福康组连续给大鼠灌胃肾福康胶囊40 d,分别按低、中、高剂量0.225、0.45、0.9 g生药/(kg·d)]进行给药;阳性对照组给予灌胃尿毒清颗粒6.7g/(kg·d))]共40 d。造模后12、24 d测定各组大鼠血清BUN、血清肌酐(SCr)含量;造模后第40天测定各组大鼠心、肾脏组织匀浆中超氧化物岐化酶(SOD)及丙二醛(MDA)含量。结果模型组大鼠血清BUN、SCr及心、肾组织匀浆中MDA含量明显高于空白组,SOD含量较空白组显著性降低(均P<0.01)。阳性对照组、肾福康中、高剂量组与模型组比较,能显著降低大鼠血清BUN、SCr及心、肾组织中MDA含量(P<0.05),明显增加心、肾组织匀浆液中SOD含量(P<0.05)。各剂量组间比较,肾福康中、高剂量组在降低大鼠血清BUN、SCr含量,升高肾组织匀浆中SOD含量方面与肾福康低剂量组比较有显著性优势(P<0.05)。结论肾福康胶囊具有抗氧化作用,能有效保护CRF大鼠心、肾等重要器官。

Abstract:Objective To observe the anti-oxidation effects of Shenfukang Capsule for rats with chronic renal failure (CRF).Methods 70 SD rats were randomly divided into blank,disease model,positive control and ShenFukang lowdose,middle-dose and high-dose treatment groups according to the levels of blood urea nitrogen (BUN),each gourp had 10 rats.All groups were fed with adenine to induce CRF model besides blank group.ShenFukang low-dose,middle-dose and high-dose treatment groups were administered according to the dose of 0.225,0.45,0.9 g/(kg ·d) respectively for 40 days.Positive control group were given NiaoDuqing Granules according to the dose of 6.7 g/(kg· d) for 40 days.The levels of BUN,serum creatinine (SCr) of all groups after molding of 12 days and 24 days were detected,and the contents of superoxide dismutase (SOD),malondialdehyde (MDA) in heart and kidney after molding of 40 days were detected.Results Compared with blank group,the levels of BUN,SCr,MDA in heart and kidney in model group were obviously higher,while the activities of SOD were obviously lower (all P 〈 0.01).The levels of BUN,SCr,MDA in heart and kidney in positive control group,Shenfukang middle-dose and high-dose treatment groups were obviously lower,the activities of SOD were obviously higher compared with model group (all P 〈 0.05).The effect of lower BUN,SCr levels and higher SOD activities in Shenfukang middle-dose and high-dose treatment groups were more significant than in Shenfukang low-dose treatment group (P 〈 0.05).Conclusion Shenfukang Capsule has the ability of anti-oxidation,can protect important organs such as heart,kidney of rats with CRF.
