绞股蓝对衰老大鼠下丘脑的抗衰老作用研究

摘    要:目的:探讨绞股蓝对衰老大鼠下丘脑的保护作用及其作用机制。方法:40只大鼠随机分为正常对照组、衰老模型组、生理盐水灌胃组、绞股蓝灌胃组。除正常对照组外均皮下注射D-半乳糖制备大鼠亚急性衰老模型。检测下丘脑中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的活性以及下丘脑谷氨酸(Glu)、一氧化氮合酶(NOS)、一氧化氮(NO)的含量。结果:衰老模型组下丘脑中SOD、GSH-Px含量明显降低;MDA、Glu、NO、NOS含量明显增高,同正常对照组比较有显著性差异(P<0.01)。绞股蓝灌胃组SOD、GSH-Px活性明显提高,MDA、Glu、NO、NOS含量降低明显,与衰老模型组比较有显著性差异(P<0.01)。结论:绞股蓝可提高抗氧化物酶的活性、抑制谷氨酸和一氧化氮的合成而发挥抗衰老效应。

Abstract:Objective:To study the effects of Gynostemma pentaphyllum Makino on hypothalamus in sub-acutely aging rats and its mechanisms.Methods:40 SD rats were randomly divideded into four groups:normal group,model group,salines group and Gynostemma pentaphyllum Makino group.Rats were injected D-galactose to make the sub-acutely aging model.The activities of SOD、MDA、GSH-Px、NO、NOS and the content of Glu in hippocampus were measured by spectrophotometer.Results:In the sub-acutely aging model group,the content of SOD、GSH-Px were lower and the activities of MDA、NO、NOS and Glu were higher than normal group(P<0.01).In Gynostemma pentaphyllum Makino group,the activity of SOD、 GSH-Px significantly increased,the content of MDA、NO、NOS decreased,and the Glu activity in the hippocampus was inhibited in aging mice compared with the normal group.Conclusion:Gynostemma pentaphyllum can restrain the activities of Glu and NO,and there is strong scavenging capacities on free radicals in aging rats's hippocampus.
