| 摘 要： | 目的 探讨芦荟大黄素(AE)与5-氟尿嘧啶(5-FU)体外协同作用及对人胃癌细胞增殖及凋亡的影响.方法 MTT法检测药物对细胞增殖抑制作用;光镜观察细胞生长状态,计数细胞分裂指数;电镜观察药物对细胞超微结构的影响;TUNEL法检测细胞凋亡.结果 AE单独应用对细胞的增殖抑制作用弱,与5-FU合用明显抑制细胞生长,呈剂量依赖性;药物作用48 h后,联合用药组细胞分裂指数显著降低,可见大量细胞皱缩,胞膜界限模糊,核膜消失,核质浓缩及凋亡小体形成;TUNEL法分析显示,联合用药组细胞凋亡率为(69.4±4.1)%,与AE组[(15.7 4±1.9)%]、5-FU组[(33.5±2.8)%]比较差异显著(P<0.05).结论 AE与5-FU联合应用能有效抑制胃癌细胞增殖,促进细胞凋亡,二药具有协同增效作用.
