交泰丸穴位贴敷对心肾不交失眠大鼠HPA轴相关激素影响的研究

摘    要:目的 探讨交泰丸穴位贴敷对失眠大鼠的催眠作用和HPA轴功能的影响。方法 取50只雄性SD大鼠分为空白对照组、模型组、穴位贴敷组、艾司唑仑组、联合干预组,每组10只。除空白对照组外,其余各实验组大鼠均采用对氯苯丙氨酸(PCPA)(450mg/kg)腹腔注射和多因素共同刺激建立失眠模型,模型建立成功后,空白对照组和模型组在进行相同时间的抓取固定,但不进行其他干预,穴位贴敷组采用0.5 cm×0.5 cm的交泰丸膏药穴位贴敷,艾司唑仑组给予0.3mg/(kg·d)艾司唑仑灌胃,联合干预组采用0.5 cm×0.5 cm的交泰丸膏药穴位贴敷后再用0.3mg(kg·d)艾司唑仑灌胃,每天 1次,连续给予14天。观察各组大鼠一般情况、体质量、戊巴比妥钠翻正实验结果,酶联免疫吸附剂测定法(ELISA)检测大鼠毛发和血清促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)和皮质酮(CORT)的水平,采用苏木素-伊红(HE)染色观察各组大鼠下丘脑组织病理改变。结果 与模型组比,穴位贴敷组、艾司唑仑组、联合干预组体质量增加(355.6±12.94)g、(347.8±22.51)g、(351.8±21.50)g比(298.1±36.45)g,p均<0.05],睡眠潜伏期减少(199.8±27.99)s、(203.2±30.65)s、(203.9±38.57)s比(251.1±49.54)s,p均<0.05],睡眠时间增加(71.7 ±13.6)min、(71.8 ±15.5 )min、(71.8 ±15.5)min比(54.9 ±5.7)min,p均<0.05]。血清中的CORT、ACTH水平显著下降,CORT:(93.99 ±3.53)(ng/L)、(94.10 ±2.78)(ng/L)、(92.05 ±3.92)比(99.73 ±1.08)(ng/L);ACTH:(140.2±1.82)(ng/L)、(140.3±2.11)(ng/L)、(138.7±1.65)(ng/L)比(144.7±1.15)(ng/L),p均<0.05]。毛发中的CRH、CORT、ACTH水平显著降低,CRH:(58.20±1.75)(ng/L)、(57.66±1.53)(ng/L)、(57.51±2.54)(ng/L)比(61.29±0.74)(ng/L);CORT:(93.99±3.53)(ng/L)、(94.10±2.78)(ng/L)、(92.05±3.92)(ng/L)比(99.73±1.08)(ng/L);ACTH:(123.0±1.70)(ng/L)、(124.7±1.36)(ng/L)、(123.3±3.43)(ng/L)比(129.8±3.52)(ng/L),p均<0.05],穴位贴敷组、艾司唑仑组合联合干预组上述指标比较差异

Abstract:Objective To investigate the effect of acupoint application of Jiao-tai-wan on hypnosis and HPA axis function in insomnia rats. Methods Fifty male SD rats were divided into blank control group, model group, acupoint application group, Esazolam group and combined intervention group, with 10 rats in each group. Except the blank control group, the other experimental groups of rats were treated with peritoneal injection of p-chlorophenylalanine (PCPA)(450mg/kg) and the combination of multiple factors to establish insomnia model. After the model was successfully established, the blank control group and the model group were treated with the same grasping fixation time, but no other intervention was performed. Acupoint application group was treated with 0.5 cm×0.5 cm Jiao-tai-wan poultice, the Esazolam group was treated with 0.3mg/ (kg·d) esazolam intragastric administration, and the combined intervention group was treated with 0.5 cm×0.5 cm Jiao-tai-wan poultice followed by 0.3mg (kg·d) esazolam intragastric administration, once a day. It was given continuously for 14 days. The general condition and body weight of rats in each group were observed. The results of pentobarbital sodium test were observed. The levels of adrenocorticotropin releasing hormone (CRH), adrenocorticotropin (ACTH) and corticosterone (CORT) in hair and serum of rats were detected by enzyme-linked immunosorbent assay (ELISA). Hematoxylin-eosin (HE) staining was used to observe the histopathological changes of hypothalamus in each group. Results Compared with model group, body mass of acupoint application group, Esazolam group and combined intervention group increased (355.6±12.94) g, (347.8±22.51) g, (351.8±21.50) g ratio (298.1±36.45) g, all p <0.05]. Sleep latency decreased (199.8±27.99) s, (203.2±30.65) s, (203.9±38.57) s vs. (251.1±49.54) s, all p <0.05]. Sleep time increased (71.7 ±13.6) min, (71.8 ±15.5) min, (71.8 ±15.5) min vs. (54.9 ±5.7) min, all p <0.05]. Serum CORT and ACTH levels were significantly decreased, CORT: (93.99 ±3.53) (ng/L), (94.10 ±2.78) (ng/L), (92.05 ±3.92) compared with (99.73 ±1.08) (ng/L); ACTH: (140.2 + 1.82) (ng/L), (140.3-2.11) (ng/L), (138.7-1.65) (ng/L) than (144.7 + 1.15) (ng/L), p < 0.05]. The levels of CRH, CORT and ACTH in hair were significantly decreased, CRH: (58.20±1.75) (ng/L), (57.66±1.53) (ng/L), (57.51±2.54) (ng/L) compared with (61.29±0.74) (ng/L); CORT: (93.99±3.53) (ng/L), (94.10±2.78) (ng/L), (92.05±3.92) (ng/L) ratio (99.73±1.08) (ng/L); ACTH: The ratio of (123.0±1.70) (ng/L), (124.7±1.36) (ng/L) and (123.3±3.43) (ng/L) was (129.8±3.52) (ng/L), all p <0.05]. There was no significant difference in the above indexes between the acupoint application group and the Esazolam combination intervention group. HE staining pathology showed that hypothalamic capillaries of rats in the model group were dilated, neurons were disordered, and vacuolar degeneration was observed. The above conditions were improved in the acupoint application group and the combined intervention group of Esazolam. Conclusion Acupoint application of Jiaotai pill may improve the occurrence and development of insomnia by regulating the function of HPA axis, prolonging sleep time and shortening sleep latency.
