人参炔醇对氧糖剥夺神经细胞损伤的保护作用

摘    要:目的:探讨人参炔醇对氧糖剥夺(OGD)致神经细胞损伤的保护作用。方法:体外培养PC12细胞,分为正常对照组、OGD组、人参炔醇组、尼莫地平组,加入含0.5 mmol.L-1 Na2SO4的无糖Earle’s液建立OGD损伤模型,以人参炔醇(10,20mg.L-1)、尼莫地平(100μmol.L-1)处理。采用荧光显微镜观察细胞形态,Hoechst和PI双染流式细胞分析仪检测凋亡率,测定培养上清液中乳酸脱氢酶(LDH)的活性与细胞Ca2+浓度,观察人参炔醇对OGD致PC12细胞损伤的保护作用。结果:人参炔醇能够减少OGD损伤PC12细胞LDH的释放(P<0.05,P<0.01),抑制Ca2+的过量产生(P<0.05,P<0.01)、显著减少细胞凋亡及坏死(P<0.05,P<0.01)。结论:人参炔醇对OGD致神经细胞损伤具有保护作用。
