运动后大鼠血清中相关酶学变化的比较研究

摘    要:目的通过检测运动后大鼠血清心肌肌钙蛋白I(cTnI)和磷酸肌酸激酶(CK),磷酸肌酸激酶同工酶(CK-MB),乳酸脱氢酶(LDH)并将它们进行比较,以期探讨它们在运动医学中的应用价值。方法38只SD大鼠分为对照组,游泳力竭后即刻组,游泳4.5h后6h,18h,24h组,分别测定其血清cTnI,CK,CK-MB,LDH。cTnI检测用改良Bodor双抗夹心酶联免疫法。CK-MB测定用免疫抑制法。结果①游泳力竭后即刻组,大负荷运动后6,18,24h组cTnI的OD值与对照组相比,t=1.567,ν=13,P=0.141;F=1.393,P=0.267,组间P值0.061~0.897,显示差异均无显著性。②游泳力竭后即刻组CK,CK-MB,LDH升高,与对照组相比,t值分别为4.027,10.666,9.413,P值均<0.01,差异有显著性。③大负荷运动后6h组CK,CK-MB,LDH升高,与对照组相比,P值分别是0.044,0.015,0.019,P值均<0.05,差异有显著性。结论①游泳力竭后即刻CK,CK-MB,LDH升高,但cTnI未有升高,尚需进一步动态检测cTnI。②大负荷运动后,动态检测未见有血清cTnI升高,提示未有心肌损伤。③大负荷运动后6h组出现的CK,CK-MB,LDH升高是否表明有心肌损伤应重新认识。

Abstract:Aim To detect and compare the variation of creatine phosphokinase(CK),MB isoenzyine creatine-phosphokinase(CK-MB), LDH and cardiac troponin(cTnI) in rats' serum pre-and post-swimming and then explorer value of serum CK, CK-MB, LDH and cTnI as an indicator of myocardial their applieation in sports medicine and sports health. Methods Thirty-eight rats were randomly divided into five groups: control group(n=7), the group of exhausted swimming(n=8), the groups studied 6(n=9), 18(n=7), 24(n=7) hours after intensive swimming respectively.By the method of immunosuppression, which used monoclonal anti-CK-M antibody, CK-MB level was detected.Serum cTnI level of each group was measured by the method of improved double monoclonal antibody Enzyme-linked immunoadsordent assay(ELISA).Levels of CK,CK-MB,LDH and cTnI in serum were all detected and compared with each other. Results Compared with the control group, the OD of the serum cTnI increased insignificantly in the exhausted swimming group and the groups 6, 18 and 24 hours after intensive swimming. But cTnI had no significant increase in all groups (t=1.567,ν =13, P=0.141; F=1.393, P=0.267). There were also no significant differences among the five groups (P=0.061- 0.897). Compared with the control group, the levels of CK, CK-MB and LDH increased significantly in the exhausted swimming group and the group 6 hours after intensive swimming (t=4.027, 10.666, 9.413, P< 0.01; P=0.044, 0.015, 0.019).Conclusion Since the level of serum cTnI does not increase in rats after intensive exercise, it is concluded that myocardial injury dose not occur. The increase in serum CK, CK-MB and LDH levels, 6 hours after intensive exercise, is not a sufficient indicator of cardiac injury. And it is possible to research cardiac injury further.
