普托品对脑缺血的保护作用

目的:探讨普托品(protopine,Pro)时大鼠局灶性脑缺血、小鼠急性脑缺血及耐缺氧的保护作用及机制.方法:采用可逆性大脑中动脉梗塞法观察Pro对局灶性脑缺血大鼠梗塞面积和行为障碍的影响;结扎小鼠双侧颈总动脉造成急性脑缺血模型观察Pro对急性脑缺血小鼠死亡率的影响;采用小鼠常压耐缺氧实验观察Pro对小鼠存活时间和耗氧量的影响.结果:Pro 60mg/kg能明显改善局灶性脑缺血大鼠神经功能及缩小脑梗塞范围;并能明显降低急性脑缺血小鼠的死亡率;Pro 40mg/kg能延长小鼠闭塞窒息生存时间,并轻度降低耗氧量.结论:Pro具有抗缺氧、抗脑缺血作用.

The protective effect of protopine on cerebral ischemia in rats and mice

Object: To investigate the protective effect of protopine(Pro,isolated from Corydalis amabilis migo, COAMTA) on anoxia and cerebral ischemia in rats and mice. Methods: The effect of Pro on neurologic deficits(ND)and infarct size(SI) in rats was studied by using the animal model of reversible middle cerebral artery occlusion (MCAO). The influence of Pro on acute cerebral ischemia was detected, which was produced by the occlusion of bilateral caroted arteries of mice. The action of Pro on anoxia was also investigated, which was produced by close normabaric hypoxia. Results: Pro 60mg/kg ip could significantly improve neurologic deficits 2h after MCAO and reduced infarct size. Pro 60mg/kg brought down mortality of acute cerebral ischemia mice. Pro 40mg/kg prolonged survial time of mice obviously and reduced oxygen consumption slightly. Condusion: These results indicated that Pro acts the part of anti-anoxia, anti-cerebral ischimia.
