人arresten因子对内皮细胞与LOVO细胞黏附的抑制

摘    要:目的:观察人arresten因子对人脐静脉内皮细胞HUVEC与LOVO细胞黏附的影响,探讨与之相关的黏附分子表达的变化。方法:半定量RT-PCR检测arresten对人脐静脉内皮细胞表达血管细胞黏附分子-1(VCAM-1)mrRNA的影响,Western-hlot检测VCAM-1蛋白表达的变化。孟加拉玫瑰红活细胞染色法观察arresten因子对内皮细胞与LOVO细胞黏附的影响。结果:RT-PCR及Western-blot结果表明VCAM-1在HUVEC细胞中高表达,但经过arresten因子处理的HUVEC细胞,其VCAM-1在mRNA水平及蛋白水平的表达均显著下降(P< 0.05)。孟加拉玫瑰红活细胞染色法显示arresten能抑制HUVEC细胞与LOVO细胞的黏附。结论:arresten因子通过降低内皮细胞VCAM-1的表达抑制内皮细胞与肿瘤细胞的黏附,这可能是其抑制肿瘤转移的一种途径。

Abstract:OBJECTIVE:To investigate anti-adhesion of HUVEC to LOVO cell induced by human arresten and measure the expression of VCAM-1 in HUVEC. METHODS: The expression of VCAM-1 in HUVEC lines was measured by semi- quantitative RT-PCR and Western blot, respectively. The adherence of HUVEC cell line to LOVO cell line was detected by using the rose bengal strain. RESULTS: The expression of VCAM-1 both at mRNA level and protein level decreased significantly in HUVEC cell line induced by human arresten (800 ng/mL)(P<0. 05). Human arresten (800ng/mL) inhibited the adherence of HUVEC cell line to LOVO cell line (P<0. 05). CONCLUSIONS: Human arresten inhibits the adherence of HUVEC cell line to LOVO cell line by inhibiting the expression of VCAM-1 in HUVEC cell line, and it may be used to prevent metastasis.
