棓丙酯对高糖诱导人肾小管上皮细胞损伤的保护作用

摘    要:目的 观察樯丙酯对高糖诱导人肾小管上皮细胞损伤的保护作用.方法 传代培养人肾小管上皮细胞(HK-2),分为对照组、高渗组、高糖组、处理组1(桔丙酯浓度2μg/ml)和处理组2(棓丙酯浓度4μg/ml),观察各组细胞增殖能力、凋亡率、活性氧(ROS)水平以及bcl-2和BaxmRNA的表达情况.结果 高糖刺激HK-2细胞后,细胞增殖能力下降、凋亡增加、ROS水平升高、bcl-2 mRNA表达减少、Bax mRNA表达增多.棓丙酯可以提高细胞增殖能力,减少凋亡,降低ROS水平,上调bcl-2 mRNA表达以及下调BaxmRNA表达;处理组2上述改变较处理组1明显(P均<0.05).结论 桔丙酯改善高糖诱导的人肾小管上皮细胞损伤,其机制可能与减轻氧化应激、上调bcl-2 mRNA表达以及下调Bax mRNA表达有关.

Abstract:Objective To observe protective effect of propyl gallate on human renall tubular epithelial cell damage induced by high-glucose and its mechanisms. Methods The renal tubular epitheli- al cells (HK-2) were divided into control group, osmolarity group, high-glucose group, treatment group 1 (2 μg/mlpropyl gallate) and 2 (4 μg/ml propyl gallate). The cell proliferation ability, rate of cell apopto- sis,intracellular level of reactive oxygen species and expression of bcl-2 and Bax mRNA were examined. Results There were no significant differences of the cell proliferation ability,rate of cell apoptosis,intra- cellular level of reactive oxygen species and expression of bcl-2 and Bax mRNA between control group and osmolarity group. Compared to control group, the cell proliferation ability were decreased, rate of cell apop- tosis and intracellular level of reactive oxygen species were increased, expression of bcl-2 mRNA were down-regulated and Bax mRNA were up-regulated. All of these changes induced by high-glucose were re- versed by treatment of propyl gallate with dose-dependence. Conclusion Prowl gallate could improve HK-2 cell injury induced by high-glucose, through attenuating oxidative stress, up-regulating the expres- sion of bcl-2 mRNA and down-regulating the expression of bcl-2 mRNA.
